Objective:Observation on the effects of runfeizhisou pill on chronicobstructive pulmonary disease (COPD) in rat models of pulmonary tissuepathology; serum interleukin-8(IL-8) and tumor necrosis factor-а(TNF-а). Discussion on mechanism of runfeizhisou pill in the treatmentof COPD.Method:30Wistar rats were randomly divided into2groups;Control Group and Create model group. Create model group is using the"smoked a joint air drops of LPS law" copy COPD rat models.Model endpathological model was successful. The Create model group is randomlydivided into model groups and treatment group. Group of rats treated byChinese herbs with traditional Chinese medicine (runfeizhisou pills) afterintragastric infusion treatment, control group and model group filled withthe ultra pure water in the stomach. Observation on three groups of rats,kill all rats after intracerebral administration intervention in30days, H-Estaining of lung tissue of the rats, observed morphological changes inlung tissue, Lung lining intervals on MLI and MAN, to estimate thedegree of emphysema in chronic obstructive pulmonary disease.Radioimmunoassay determination of content of serum TNF-а and IL-8in rats in each group. Using SPSS13statistical software to analyze theresults.Results:1Runfeizhisou pill can improve the mental status and coughingexpectoration, wheezing and other symptoms of rat COPD models.2Three groups of pathological changes in lung tissue of rats Lungtissue of rats with control group no significant inflammatory cellinfiltration, bronchial mucosa without thickening and bronchial epithelial Cilia arranged in neat, no rupture of the thin alveolar wall, no pulmonaryBulla formation. Model groups shows inflammatory cell infiltration inlung tissue of rats, mainly macrophages, lymphocytes and mononuclearcells, bronchial wall thickening, hair shedding and visible mucous inthrombus formation and chronic airway inflammatory cell infiltration andalveolar walls form a ruptured bullae, thickening of blood vessels arevisible. Mainly changes of Lobular Central type pulmonary emphysemain model group. Treatment group shows model group’s changes, modelgroup of chronic inflammatory cell infiltration degree, emphysemadamage lighter than the model group.3Comparison of three groups of morphology of lung tissue in rats3.1Comparing the MLI in three groups of rats.Control rat MLI on average:(8.25±0.59)×10-5m, modelgroup is (13.27±1.55)×10-5m, treatment group is (11.2±0.84)×10-5m. Through statistical analysis, control group is better than thetreatment group, treatment group over model group. Model groupsstatistically significant difference compared with the control group(P<0.05), description model groups increases rat alveolar mean diameterthan control group; Treatment group compared with the model groupsstatistically significant difference (P<0.05), alveolar mean diameter lessthan the treatment group model group; Treatment group compared withthe model groups, statistically there is a significant difference (P<0.05),indicating alveolar treatment group average diameter is larger than thecontrol group. After runfeizhisou pill treatment, treatment group alveolarmean diameter less than the model group, but still larger than the controlgroup.3.2Comparison of three groups of MAN ratsControl group lung mean alveolar number is (11.38±2.32)×107/m2, model group (5.04±0.88)×107/m2, treatment group (6.79±1.17)×107/m2. The control group rat’s lung tissue mean alveolarnumber is more than model group, statistically there is a significant difference (P<0.05), the density of alveolar of rats in the model groupcompared with control group decreased; Treatment of lung tissue of ratsmean alveolar number compared with the model group, less than thecontrol group, statistically significant differences (P<0.05), explain thetreatment group after treatment in rat alveolar density higher than themodel group, lower than that of the control group.Morphological Comparison of three groups of rat’s lung tissueindicates, COPD model rats with pulmonary emphysema. After thetreatment, the treatment group rats emphysema extent lighter than that ofmodel group rats, but still in control rats.4Three groups of rat’s serum TNF-а comparisonControl rat serum TNF-аnumber is (0.67±0.57)ng/ml, themodel group (1.22±0.15)ng/ml, the treatment group (0.83±0.08)ng/ml. Elevated TNF-аlevels compared with the model group and thenormal group, the treatment group compared with the model group, theTNF-level decreased, increased TNF-levels compared with traditionalChinese medicine treatment group and the normal group, the three groupof two statistical significant difference (P<0.05). Indicates that COPDmodel group rats TNF-а in a high level state,the runfeizhisou pilltreatment can reduce the rat model of COPD TNF-а levels, but not tothe level of the control group.5Three groups of rat’s serum IL-8comparisonControl rat serum IL-8number is (0.22±0.06) ng/ml,the modelgroup (0.49±0.08)ng/ml,the treatment group (0.33±0.03) ng/ml.Comparison of serum IL-8level increased in model group rats andcontrol rats,statistically significant differences (P<0.05), serum IL-8of ratmodel of COPD in a high level state;the treatment group compared withthe model group,the serum IL-8level was decreased,with significantdifference (P<0.05),elevated IL-8levels compared with control serumtreated group group,statistically significant differences (P<0.05),indicating that after runfeizhisou pill treatment can decrease the IL-8 level in serum of COPD rat model,but not to the level of the controlgroup.Conclusion:1Runfeizhisou pill can increase the clinical symptoms ofCOPD rats.2Runfeizhisou pill can relieve the inflammatory reaction in thelung tissue of COPD rat model, slow or prevent progression ofemphysema.3Runfeizhisou pill can decrease the COPD model rat’s serumTNF-а, IL-8levels. To alleviate the pulmonary inflammatory reaction,so as to achieve the therapeutic effect on COPD.
