Objective:To explore the influence of the cytokines secreted by bone marrow mesenchymal stem cells (BMSCs) and embryonic stem cells (ESCs) on delayed type hypersensitivity (DTH) reaction and provide the experimental basis for the clinical application of the two kinds of cytokines as a new treatment to allergic diseases.Methods:1. Mouse BMSCs were isolated from mouse bone marrow and purified by adherence screening in the aseptic conditions. The stem cell cytokines from BMSCs were obtained from the supernatants of third passage BMSCs at cell density reached80%to90%. ESCs were obtained from the primordial germ cells in mice embryonic gonadal ridges and then developed and extended. ESCs cytokines were obtained by the same way as BMSCs cytokines.2.Mice were randomly divided into normal control group, model group, medium group, dexamethasone group, BMSCs cytokines group and ESCs cytokines group. Mouse model for allergic contact dermatitis (ACD) in five groups was established by sensitization by applying0.5%dinitrofluorobenzene (DNFB) on the shaved area of mice abdomen and evocation on its ears (normal control group doesn’t be handled) to observe the effects of cytokines from BMSCs and ESCs on the auricle swelling test, thymus and spleen index with dexamethasone as the positive control.3. Prepare paraffin section of the line HE staining of each group of mice auricles to observe the histological differences and the changes of the auricle epidermis thickness and total thickness of mice auricle with microscope.Results:1. Compared with model control group, dexamethasone and the cytokines from BMSCs and ESCs could obviously relieve auricle swelling degree, red ice thymus and spleen index, and there were all significant differences(P<0.01). Less inflammatory cells infiltration, auricle epidermis thickness and the total thickness were observed with light microscope(P<0.05).2.The influences of the cytokines from BMSCs and ESCs were waker than that of dexamethasone (P<0.05).3.The effect of cytokines from BMSCs was waker than that of cytokines from ESCs(P<0.05).Conclusions:These demonstrated that cytokines from BMSC&and ESC&had the inhibitory effect on DTH, however,the inhibited effect on DTH of cytokines from ESCs more obviously than that of cytokines from BMSCs.
