OBJECTIVE:To explore the expression of the liver X receptor (LXR) and the correlative of cholesterol ester transfer protein (CETP), cyclooxygenase-2(COX-2) in school-age children with obesity. And to explore the relative factors of liver X receptor (LXR) and lipid metabolism in school-age children with obesity.METHODS:The pupils in Grades1-6, aged7-14were selected from8primary school of Lanzhou during June to October in2011. By detecting students’ BMI,80students with obesity were selected as experiment group. Based on the age and sex matched method,51ordinary children were selected as control group. Fasting blood were used to detect the expression levels of metabolic indexes, to declutch and culture macrophage, from which RNA was extracted. Real time fluorescence quantitative PCR was used to detect the relative expression levels of LXR, CETP and COX-2.RESULTS:The expression levels of LXR, CETP and COX-2in experiment group were higher than control group, there were statistic difference between the two groups (P<0.05). There was no statistic difference between boys group and girls group in the expression levels of LXR, CETP and COX-2(P>0.05). COX-2and CETP were positively correlated with LXR respectively (P<0.05). The levels of AST、ALT、GGT、CHOL、TG and LDL-C in experiment group were higher than that in control group, and HDL-C was lower, there were statistic difference between the two groups (P<0.05); The correlation analysis found that, AST、ALT、CHOL、LDL-C and BMI were positively correlated with liver X receptor (r=0.18~0.26, P<0.05). Logistic regression analysis showed that high AST, ALT, CHOL, LDL-C and BMI were risk factors in the increasing of LXR (P<0.05). Another logistic regression analysis showed that ST≥40IU/L (OR=1.076), ALT≥40IU/L (OR=1.036), CHOL≥5.20mmol/L (OR=2.038), LDL-C≥3.36mmol/L (OR=2.176)、BMI≥18.9kg/m2(OR=1.131) were risk factors in the increasing of LXR (P<0.05).CONCLUSION:Obesity could lead to a higher expression of LXR, CETP and COX-2in school-age children; The expression of LXR, CETP and COX-2in school-age children are unaffected by gender; Obesity on school-age children can upregulation of the expression of liver X receptor and cause liver damage and lipids disorders.
