AIM: To explore the effect of Toll-like receptor2and4in mycobacteriumbovis Bacillus Calmette-Guerin (BCG) induced human proximal renal tubuleepithelial cells injury. METHODS: HK-2cells were stimulated by BCG, theexpression of TLR2, TLR4, chemokine (C-X-C motif) ligand1(CX3CL1),transforming growth factor beta1(TGF-1),E-cadherin(E-cad),α-Smooth muscleactin(α-SMA)was detected by quantitative real-time PCR or Western blot. TLR2monoclonal antibody and TLR4inhibitor were treated with HK-2cells1h beforeBCG stimulation. The expression of CX3CL1, TGF-1,E-cad and α-SMA wasevaluated by quantitative real-time PCR or Western blot. RESULTS: BCGincreased the expression of TLR2, TLR4, CX3CL1,TGF-1,α-SMA anddownregulated the expression of E-cad in HK-2cells.The expression of TLR2 and TLR4was in time-dependent manners and dose-dependent manners, theexpression of CX3CL1,TGF-1,α-SMA and E-cad was in time-dependent manners.Additionally, the expression of CX3CL1,TGF-1,E-cad and α-SMA inhibitedpartly by TLR2monoclonal antibody and TLR4inhibitor respectively.CONCULUSION: BCG is able to increase the productions of TLR2, TLR4,CX3CL1,TGF-1,α-SMA and decrease the productions of E-cad in HK-2cells.TLR2and TLR4signaling pathways play important roles in tubule epithelial cellinjury induced by BCG.
