Objective The aim of this study is to detect the effect of Pluronic F-127hydrogelcombined with stromal cell derived factor-1α(SDF-1α)on bone mesenchymal stemcells(BMSCs)in rats’s chemotaxis in vitro,and to observe the biocompatibility ofBMSCs cultured in Pluronic F-127gel, in addition to explore the possibility of BMSCsdifferentiating into osteoblast three-dimensionally cultured in SDF-1α-gel。Method Transwell chamber was used to detect the effect of Pluronic F-127hydrogelcombined with various concentration of SDF-1α (200,50ng/ml) on BMSCs’smigration,the number of the transfer cells can be statistical analysis by the results of theexperiment,and the better composite hydrogel can be selected. MTT was used toexplore the cytoactive of BMSCs after cultured in the chosen SDF-1α-gel. The BMSCswas osteogenesis cultured in3-dimensional scaffold SDF-1α-Pluronic F-127hydrogel,at the same time respectively put the simple BMSCs cultured in osteogenesis andcomplete medium,then alkaline phosphatase (AKP) activity and alizarin red wasevaluated for each of them,at last recorded the results and analysis the osteogeneticdifferentiation of BMSCs.Result The result of chemotaxis display that the concentration of SDF-1α200ng/ml inhydrogel has obvious effect on migration of BMSCs compared to the concentration ofSDF-1α50ng/ml, the transfer cells number were increased significantly on the surfaceof membrane filter below(P<0.05),MTT confirmed that PluronicF-127have no effecton cell proliferation(P>0.05). Compared with control group, BMSCs-gel group and pure BMSCs group’s AKP have significantly increased in the circumstances ofosteogenesis induction(P<0.05), and calcium nodule can be found in the inductiongroup, however there were no calcium nodule formation in the control group.Conclusion The composite material PluronicF-127loaded with SDF-1α weresignificantly promote chemotactic migration of BMSCs in vitro, BMSCs were able tosurvive normally,grow and proliferate very well in PluronicF-127,BMSCs can beinduced differentiation into osteoblast in the3-dimensional scaffold SDF-1α-gel.
