Objective To study the expression of TSLP、TSLPR and NF-κB in the endometrial glandular cell and stromal cell of adenomyosis patients,as well as the serum level of TSLP, and to vestigate the role of TSLP、TSLPR and NF-κB in the pathogenesis of adenomyosis.Methods Immunohistochemistry was employed to detect the expression of TSLP, TSLPR and NF-KBp65in the endometrial glandular cell and stromal cell of35adenomyosis patients and20hysteromyoma patients as control, and analyze the correlation of the two group protein TSLP and NF-κBp65, TSLP and TSLPR. The concentration of TSLP was measured by specific ELISA in the serum of20healthy subjects besides of the patients with adenomyosis and hysteromyoma.Results TSLP、TSLPR and NF-KBp65were both positively expressd in the glandular cells and stromal cells of ectopic and eutopic endometrium of patients with adenomyosis, their expression was significantly higher than that of the control group (P<0.01), and the expression of TSLP、TSLPR and NF-KBp65in ectopic endometrium was significantly higher than that in eutopic endometrium (P<0.01). There was positive correlation between TSLP and NF-KBp65in ectopic endometrium. But there was no obvious correlation between TSLP and TSLPR in the glandular cells and stromal cells of ectopic and eutopic endometrium of patients with adenomyosis. The concentration of TSLP was higher in the serum of patients with adenomyosis.Conclusion (1) The expression of TSLP、TSLPR and NF-κB in eutopic endometrium of patients with Adenomyosis was higher than that in control group, and it illustrates that there was speciality in eutopic endometrium deffering from the normal endometrium, thus reflecting and proving "the determinism of eutopic endometrium";(2) The expression of TSLP、TSLPR and NF-KBp65in ectopic endometrium was significantly higher than that in eutopic endometrium, and it points out that the form of the ectopic endometrium has relation with the expression of TSLP、TSLPR and NF-κB. And besides, TSLP and TSLPR may promote the proliferation of the endometrial cells by postponing the apoptosis of the ectopic endometrium or autocrine, moreover cause adhesion and invasion of the ectopic endometium; TSLP and TSLPR maybe act on the target cells and produce inflammatory cytokine, then indirectely promote the Th2cellular immunity, thus causing the local inflammation, which accelerate the development of ectopic endometrium;(3) TSLP and NF-κB were both highly expressed in the glandular cells and stromal cells of ectopic and eutopic endometrium of patients with adenomyosis, and there was positive correlation between TSLP and NF-KBp65in ectopic endometrium, which indicates that the overexpression of the TSLP has connection with the activation and overexpression of the NF-κB.
