Objective: To detect the expression of ki-67mRNA and Caspase-3 mRNA in glandular epithelium cell of endometrium in adenomyosis ,leiomyoma(control group),and to detect the expression of them in glandular epithelium cell of endometrium after treatment with mifepristone in adenomyosis. To explore the proliferation and apoptosis in the glandular epithelium cell of endometrium in adenomyosis , mifepristone,and control group.Methods: The endometrial cell of adenomyosis and leiomyoma was isolated and purifed by enzymolysis,filtration,and pasted wall purification. The glandular epithelium cell of endometrium was identified by immunocytochemistry.To seed logarithmic phase cell of glandular epithelium cell of endometrium with adenomyosis to 96well-multiwell plate,and randomly divide it into control group,disease group(,10-6,10-5,10-4,10-3 mol/L mifepristone)mifepristone group.After having cultured glandular epithelium cell of endometrium for 48 hours,to detect the vigor of cell by MTT,and to screening effective drug concentration of mifepristone.To culture glandular epithelium cell of adenomyosis, leiomyoma uteri,to treat the glandular epithelium cell of endometrium with adenomyosis by mifepristone ,and to design them as disease group, control group, mifepristone group ,and to analysis the expression of Caspase-3 mRNA and Ki-67mRNA in the above-mentioned three groups by semi-quantitative RT-PCR.Results: Both of proliferative and secretory phase ,the expression of Ki-67mRNA in disease group is stronger than control group(P﹤0.05).The expression of Ki-67mRNA is decreased in disease group (P﹤0.001). In proliferative phase ,the expression of Caspase-3mRNA in disease group is weaker than control group (P﹤0.01) .But in the secretory phase ,the expression of Caspase-3mRNA in disease and control group is not different (P>0.05). In proliferative phase, the expression of Caspase-3 mRNA in mifepristone group is stronger than disease group(P﹤0.05).But in secretory phase, the expression of Caspase-3mRNA in mifepristone group and disease group is not different(P﹥0.05).Conclusion: The expression of ki-67mRNA in disease group is stronger than control group, and the expression of Caspase-3mRNA in disease group is weaker than control group. The disturbanlance of apoptosis and proliferation in the glandular epithelium cell of endometrium with adenomyosis maybe one pathogenesy of adenomyosis. Mifepristone can make the expression of Caspase-3mRNA stronger in proliferative phase mifepristone group than disease group,and make the expression of ki-67mRNA in mifepristone group weaker than disease group .the mifepristone drug maybe induce the apoptosis of the glandular epithelium cell of endometrium, and can inhibite the proliferation of the cell.Then it may hold the developing of the adenomyosis.
