Objective:To investigate effect of acupuncture on expression of TNF-a、TGF-β1in oviduct tissues and IL-1β、IL-2、IL-6in serum of the Tubal Obstructive Infertility rats. To explore the molecular mechanisms of prevention and treatment of Tubal Obstructive Infertility with acupuncture. To prove the importance of acupuncture dialectical theory in prevention in Tubal Obstructive Infertility.Methods:A total of60female Wistar rats are randomized into4groups with15rats in each. They are control group, model group, acupuncture group and medicine group. To copy the phenol mucilage stimulation that injects from uterine bifurcation direction towards the ovary in Tubal Obstructive Infertility rats model. Modeled after21days, control group and model group are given the same amount of distilled water every day. The medicine group is given drug drench according to the conversion operator according to human and rat body surface area. The acupuncture group is given acupuncture30min every day. After4weeks’treatment, dissect the mouse and take out the oviduct organization and blood in the abdominal aorta. Serum and organization are preserved in the low temperature refrigerator.Immunohistochemistry method is used to observe IL-1β、IL-2、IL-6in serum to reflect the expression of inflammatory factors and the severity of the systemic inflammatory response in each group. Reverse transcription PCR method is used to observe expression of Tumor necrosis factor (TNF-a). Expression of TNF-amRNA in the oviducts at mRNA levels is detected by RT-PCR to reflect the regulation of acupuncture on TNF-amRNA and influence of local tissue inflammation trend. Wersten blot method is used to observe expression of Transforming growth factor-betal (TGF-β1) to reflect the trend in the oviduct tissue proliferation, fibrosis and obstruction.Results:The model group has the obvious oviduct organization inflammation response and the obstructive oviduct channel phenomenon. The acupuncture group and the medicine group have respective degrees of positive change comparing with the model group. There is no statistical difference between medicine group and acupuncture model (p>0.05). Immunohistochemistry method is used to observe IL-1β、IL-2、IL-6in serum of the levles in Tubal Obstructive Infertility model.The expression of model group is increased than control group (p<0.01). The expression of medicine group is decreased than the control group (p<0.01). Acupuncture group is decreased than the control group (p<0.01), but medicine group and acupuncture group have no statistical significance (p>0.05). Acupuncture does have positive effects for the treatment of oviduct obstructive infertility.
