| 1ObjectiveBy observing the influence of the Tong Du Tiao Shen acupuncture method ondynamic change of TNF-α and CRP level of focal cerebral ischemia reperfusion modelrats, to master various relevant datas in each period of the acupuncture treatment onfocal cerebral ischemia reperfusion rats and deduce the optimal time point of theacupuncture therapy.2Methods72adult Wista male rats were randomly divided into four groups, the blank group,control group, model group and acupuncture group. Each group was divided into threesubgroups by ischemia reperfusion time6h,24h,48h, six in each group. Didn’t do anydeal with blank group with normal husbandry; Control group did total artery separation;The acupuncture group and model group rats were built middle cerebral arteryocclusion (MCAO) of ischemia reperfusion model by using middle cerebral artery linebolt; Acupuncture group, by applying the method of Tong Du Tiao Shen, choseacupuncture points of BAIHUI, DAZHUI, FENGFU at corresponding time points toacupuncture. After the success of the building, on the basis of Zea first Longa nervefunction score (neurological severity score, NSS) rating standard, model rats whichconformed to the corresponding standard were included and those were eliminated andsupplemented which did not conform; Collectting blood from abdominal aorta andtakeing brain tissue by Beheadeding to detect the levels of TNF-α, CRP in serum andbrain tissue of rats with using the double sandwich ABC-ELISA method.3Results(1) Zea Longa nerve function score;(2) TNF-α, CRP levels in brain tissue and serumof control group compared with blank group had no significant difference (P>0.05);(3)TNF-α, CRP level in brain tissue and serum of model groups (ischemia reperfusion6h, 24h,48h) compared with blank group and control group were significantly higher (P <0.01). Among them, TNF-α, CRP levels in brain tissue and serum were to peak at theischemia reperfusion6h.(4) The levels of TNF-α, CRP in brain tissue and serum ofacpuncture group(ischemia reperfusion6h,24h,48h)and model group(ischemiareperfusion6h,24h,48h) had significant difference at the same point in time(P <0.01).At the same time acupuncture group in different time periods in the brain tissueand serum TNF alpha, CRP levels are compared significant difference (P <0.01), andthe ischemia reperfusion6h were the most significant (P <0.01).4Conclusion(1) Tong Du Tiao Shen acupuncture method could significantly relieve nervefunction defect of cerebral ischemia reperfusion model rats.(2) Tong Du Tiao Shen acupuncture method could reduce TNF-α, CRP levels inbrain tissue and serum of cerebral ischemia and reperfusion model rats.(3) Cerebral ischemia reperfusion model rats6h was the best time point fortreatment with Tong Du Tiao Shen acupuncture method, which could maximizelyreduce inflammation associated with cerebral ischemia reperfusion model in rats. |
