Objective:Prepared by modified Zea-Longa cerebral ischemia-reperfusion model(Middle Cerebral Artery Occlusion model/Reperfusion model,MCAO/R model). To observe the effect of three drug treatments as Tong Du Tiao Shen acupuncture and moxibustion, conventional acupuncture and Tongxinluo drug on the expression levels of cortex CD34,CD133 in ischemic area and serum VEGFR-2 of rats with MCAO/R and compare the difference of effect of the three treatments.In this study, I planed to explore the role of Tong Du Tiao Shen acupuncture and moxibustion on rat cerebral protection and angiogenesis in MCAO/R model.Method:This study choose 200,male SD rats weighing 275±25g.After one week adaptive feeding,30 rats were randomly selected as the sham group(group A),and the remaining rats as the model by modified Zea-Longa method.To exclude the unsuccessful models and to select and divide successful and behavioral score 120 ones into four groups with the computer random method:Tong Du Tiao Shen acupuncture and moxibustion group(group B),conventional acupuncture group(group C),Tongxinluo drug group(group D)and model contrast group(group E),with each group of 30 rats and being grouped treatment.Group B were treated by Baihui moxibustion for 30 min for each time,Dazhui bloodletting with 1-3 drops for each time;Group C had an acupuncture treatment with Fengchi,Quchi,Hegu,Zusanli,Sanyinjiao;Group D were forced to eat Tongxinluo capsules for an avage of 1.0g / kg.d each time;Group A and E were untreated.Group B,C and D were treated 1 time every day and total of 28 days.Before and at the end of all courses,to record neurobehavioral scores for each group with the improved method Bederson respectively.On the 7th,14 th,21th and 28 th day of treatment,5 rats were selected from each group to be collected 5ml blood under sterile conditions from abdominal aorta.To sacrifice the rats and take out the brains immediately.To slice thebrain sections embeddied with paraffin and test the positive expression of CD34,CD133 in ischemia cortical area by immunohistochemistry.The serum was tiseparated from blood samples,using ELISA method to examine the activity of VEGFR-2.To collect the specienion form each group and do index detecting.To test and analysis the results with statistical software SPSS17.0.Results:(1)Before and after the treatment: the neurobehavioral scores of all groups were both lower.Compared with group A,there was statistically significant difference in group B,C, D, E(P<0.01);Compared with group E,there was statistically significant difference in group B,C,D(P<0.01);Compared with group D,there was statistically significant difference in group B and C(P<0.01);Compared with group C,there was statistically significant difference in group B(P<0.01).The results showed that: the rats’ neurobehavioral scores of group B,C,D were reduced to varying degrees,and group B was better than group C and D(2)From7th day to28 th day,the expression quantity of CD34,CD133 in ischemia cortical area and serum VEGFR-2 decrease gradually for group B, C, D, E,the change of group A and E had no statistical significance(P>0.05).Compared group A,B,C,D,E in the 7th,14 th, 21 th,28th day of four time points respectively:The 7thd:Compared with group E,there was statistically significant difference in group B, C, D,(P<0.01);Compared with group D,there was statistically significant difference in group B and C(P<0.01,P<0.05);Compared with group C,there was statistically significant difference in group B(P<0.01);The 14thd:Compared with group E,there was statistically significant difference in group B, C, D(P<0.01);Compared with group D,there was statistically significant difference ingroup B,C(P<0.01);Compared with group C,there was statistically significant difference in group B(P<0.01);The 21thd:Compared with group E,there was statistically significant difference in group B,C,D(P<0.01);Compared with group D,there was statistically significant difference in group B,C(P<0.01);Compared with group C,there was statistically significant difference in group B(P<0.01);The 28thd:Compared with group E,there was statistically significant difference in group B, C, D(P<0.01);Compared with group D,there was statistically significant difference in group B,C(P<0.01);Compared with group C,there was statistically significant difference in group B(P<0.01).The results showed that:on one hand:After cerebral ischemia-reperfusion,the expression of CD34,CD133 in ischemic cortex area and serum VEGFR-2 of rats was higher at the7 thd,and the expression was reduced;On the other hand:There is some intervention of Tong Du Tiao Shen acupuncture and moxibustion,the routine acupuncture,Tongxinluo drug on the expression levels of CD34,CD133 in ischemic cortex area and serum VEGFR-2 of rats with MCAO/R,and Tong Du Tiao Shen acupuncture and moxibustion promotes CD34,CD133,VEGFR-2 expression effect is better than that of routine acupuncture and tongxinluo drug.Conclusion:(1)There is some improving effect of Tong Du Tiao Shen acupuncture and moxibustion,the routine acupuncture group,Tongxinluo drug on neurobehavioral scores of MCAO/R rat model.Tong Du Tiao Shen acupuncture and moxibustion is better than routine acupuncture group and Tongxinluo drug group.(2)There is some intervention of Tong Du Tiao Shen acupuncture and moxibustion, the routine acupuncture,Tongxinluo drug onthe expression levels of CD34,CD133 inischemic cortex area and serum VEGFR-2of MCAO/R rat model and.Tong Du Tiao Shen acupuncture and moxibustion is better than routine acupuncture and Tongxinluo drug.(3)Tong Du Tiao Shen acupuncture and moxibustion is effective in promoting the recovery of neurological function in MCAO/R rat model,which may be the mechanism to mobilize bone marrow stem cells homing to ischemic areas and promote angiogenesis in the ischemic area.
