Rhizoma Dioscoreae Nipponicae is the dry rhizoma of Dioscorea nipponica Makino which is a plant of Dioscoreae and it is used as a Traditional Chinese Medicine which is also known as Chuandilong, Chuanlonggu and so on. It can exspell pathogenic wind-wet, remove blood stasis, clear thread, clear up lung wet-heat and eliminate wet sputum, for clinical treatment of rheumatoid arthritis, chronic bronchitis, wounds, cardiovascular disease, etc. It is widely distributed in Northeast, Heilongjiang, Jilin, Liaoning, Inner Mongolia and other provinces. Pharmacologically and clinically, it was reported that total saponins in it had an obvious anti-inflammatory and analgesic, anti-tumor activity, vasodilating effect, stopping cough and quelling asthma effects. Besides, diosgenin in it is one of the main raw material for synthetic steroid hormones, so its medicinal value is very high.In this thesis, the chemical constituents of 75% EtOH extract from Rhizoma Dioscoreae Nipponicae were studied.6 compounds were isolated by AB-8 macroporous resin column, silica gel column and repeated recrystallization. 1 compound(aglycone) was isolated by acid hydrolysis, extraction and recrystallization. On the basis of the chemical and spectral methods, the structures of these 7 compounds were identified as ProgeninⅡ(1), Dioscin(2), Gracillin(3), Methylprotodioscin(4), Methylprotogracillin(5),β-sitosterol(6), Diosgenin(7).On the basis of 2005th year China Pharmacopoeia and references in the literature, the characters, micro structure, TLC identifications and extracts inspection were done, and a RP-HPLC method for determination of diogenin was established. In order to more effectively control the quality of Rhizoma Dioscoreae Nipponicae, for the first time, we established a method for determination of total saponins by colorimetry and a RP-HPlC method for the simultaneous determination of dioscin and gracillin.And the contents of the total saponins in different places were compared. Based on the above research, the preliminary study of HPLC-FPS analysis were done.In this paper, studies on the contents of active ingredients in Rhizoma Dioscoreae Nipponicae of different ages, different harvest seasons, different drying methods were carried out.We examined extraction conditions of total dioscorea saponins by indicators of saponins and extraction efficiency. And based on the study of extraction prodess, research of the macroporous resin purification.technology were carried out. The tablet was preparated and its quality control was studied.These results can be served as basement for the further study of Dioscorea nipponica Makino. They also afford reliable experimental basis for the application of HPLC in quality control of Traditional Chinese Medicine.
