Objective To observe the effect of Dioscorea nipponica Makino on 3T3-L1 adipocytes through the glucose intake and the expression of nuclear factor-kappaB and to detect the mechanism of the improvement of insulin resistance and the role of inflammatory response.Methods The international method was used on 3T3-L1 adipocytes differentiating to mature adipocytes and the oil red O staining was used to identify the cell differentiation. After cell count,the ceils were individed into 6 groups as normal group,model group, positive control group(rosiglitazone) and Dioscorea nipponica Makino groups(high, medium and low concentration).All of the groups were dealed with insulin(10μmol / L) or without insulin.At 48h of the drugs intervention,the 2 - deoxy -[3H]-D-glucose uptake rate of 3T3-L1 adipocytes were tested.At the same time,the nuclear protein was extracted and the Electrophoretic Mobility Shift Assay was used to detecte the expression of nuclear factor-kappaB.Results Compare with the model group,the glucose uptake rate of 3T3-L1 adipocytes in Dioscorea nipponica Makino groups(high,medium and low concentration) increased (P<0.05).There were no significant differences in Dioscorea nipponica Makino groups (high,medium and low concentration) and the positive control group(P>0.05).The expression of the nuclear transcription factor NF-κB in Dioscorea nipponica Makino groups(high,medium and low concentration) decreased(P<0.05).There were no significant differences in Dioscorea nipponica Makino groups(high,medium and low concentration) and the positive control group(P>0.05).Conclusions Dioscorea nipponica Makino increase the glucose uptake rate of 3T3-L1 adipocytes.Dioscorea nipponica Makino inhibit the expression of nuclear factor-kappaB. Dioscorea nipponica Makino may improve the role of insulin resistance,through the inhibition of adipocytes in the nuclear transcription factor NF-κB activation and inhibiting the inflammatory response to achieve. Objective To observe the therapeutical effect of Dioscorea nipponica Makino on rats with acute gouty arthritis triggered by the crystallization of uric acid.Methods 56 rats were randomly divided into 7 groups as Dioscorea nipponica Makino groups(A,B,C,D),Colchicine group,acute gouty arthritis model group and normal group.At 2 h,4h,6h,8h,10h,24h,48h and 72 h after the establishment of acute gouty arthritis model,joint-drive swelling and activities were observated.At 72 h after the establishment of acute gouty arthritis model,white blood cell count in plasma was observed and serum interleukin-1βwas detected.Results After the establishment of acute gouty arthritis model,the joint swelling increased in each group and reached a peak at 8~12 h.The degree of joint swelling decreased gradually from Dioscorea nipponica Makino low dose group to high dose group and there was significant difference between these Dioscorea nipponica Makino groups and acute gouty arthritis model group.Among those groups,high dose group decreased the most.White blood cell count in plasma decreased in Dioscorea nipponica Makino groups, comparing with the acute gouty arthritis model group.Serum interleukin-1βdecreased in Dioscorea nipponica Makino groups,comparing with the acute gouty arthritis model group.Conclusions The mid-dose and high-dose Dioscorea nippomca Makino can effectively control acute gouty arthritis attack and the effect is dependent on dose.Dioscorea nipponica Makino may inhibit the formation of leukocyte and the secretion of IL - 1β.
