Objective:To investigate the treatment effect of Dioscorea nipponica Makino-extracts on metabolism indicators in the obesity rats.Methods:8 SPF pregnant rats,96 new-born rats,the baby rats were divided into two groups on the second days,one group is called MSG model group with subcutaneous injection of L-glutamate 4g/kg.d;the other is called normal control Group with saline injection.Seven consecutive days.On the time of 21d they were weaned.Regularly record body weight,observed body-type changes.The two groups have been fed with normal diet.20w-old,chose weight gain larger male rats about 30 from MSG group and then they were randomly divided into Dioscorea nipponica Makino total saponin group, Wendiya group,model control group,the normal group with seven rats altogether in this study.Record body weight and body length,calculating Lee's index,took blood from epicanthus venous plexus to detect the level of high-density iipoprotein cholesterol (HDL-C),total cholesterol(TC),triglycerides(TG),insulin(Ins),free blood glucose(FBG) over night.Dioscorea nipponica Makino Group were gavaged with Dioscorea nipponica Makino total saponin+ DMSO,Wendiya Group Wendiya + DMSO,model control group and normal group were given distilled water + DMSO,to the total time for 8 weeks,measuring body weight,body length,and calculated Lee's index again.Also HDL-C,TC,TG,Ins,BG were detected again.After that all rats were killed,weight fat tissue aroud the renal and retroperitoneal.Preserved liver,kidney,pancreas,adipose tissue in liquid nitrogen,and collected the liver,pancreas to 10%neutral formalin-fixed. HE staining for the liver,Mallory staining for the observation of pancreatic tissue.Results:①Compared to normal group,Lee's index difference of 16-week-old MSG rats was significant(P<0.01).When the rats are 20 weeks old,biochemical markers including HDL-C,TC,TG,Ins,BG and Lee's index between MSG group and normal group were significantly different(P<0.01)②Before and after treatment,there were significantly difference between Dioscorea nipponica Makino total saponin group and Wendiya group in HDL-C,TG,Ins,BG(P<0.01),but TC change in Wendiya group was not significant(P>0.05),whereas in Dioscorea nipponica Makino Group it was significant(P<0.05).TG,Ins,BG in model control group also increased significantly (P<0.01),HDL-C,TC have no significant changes.HE staining results for liver observation showed that Dioscorea nipponica Makino total saponin can improve fatty liver,almost to normal.Wendia also improved,but not the same as Dioscorea nipponica Makino.Compared Wendiya to Dioscorea nipponica Makino total saponin group,there were no significant differences in biochemical markers after gavaging(P>0.05).Conclusion:Dioscorea nipponica Makino total saponin can reduce INS,TC,TG, FBG,increase HDL-C,and lower IRI in obesity rats.And we found total saponin has a better effect in reducing TC than Wendiya,liver HE staining showed that it can significantly improve the fatty liver disease,and is also superior to Wendiya.This study open up a new pathway to study the effective part of Dioscorea nipponica Makino in insulin resistance cure.
