As one of the rapid development of new chromatographic techniques, Capillary electrophoresis (capillary electrophoresis, CE), which is extremely suitable for protein separation and analysis, has received widespread attention due to superior characterization of high separation efficiency, short analysis times and low sample and reagent consumption. However, the absorption of proteins by the inner wall of capillary has become a problem for the separation of proteins. Nowadays, it has become a most important and commonly used method to inhibit protein adsorption by coating on the inner wall of capillary. According to whether chemical bonding will happen or not, it could be subdivided into physical adsorption coating and chemical bonded coating, which have advantages and disadvantages to both approaches. It has become a key issue of how to combine the advantages of both, preparing capillary electrophoresis coating with high quality and high performance.In this paper, layer self-assembly (LBL) technology was employed to establish structure controllable of self-assembled thin films on the inner wall of capillary, by replacing toxic silane reagent water-soluble photosensitive polymer. Under the effect of ultraviolet irradiation, in situ crosslinking reaction was induced by photosensitive polymer, thereby forming a covalently bonded self-assembled coating on the capillary wall. The separation of protein mixture using the coating has gained better results. The main work is as follows:1. DR-(PVSA) multilayer films were formed by hydrolysis product of polyvinylpyrrolidone (PVP) through the principle of layer-by-layer self-assembly. At the same time, ionic bonds converted into covalent bond under ultraviolet irradiation, forming multilayer covalently bonded coating through repeated cycles. The coating on a variety of basic proteins can achieve baseline separation and good reproducibility as well as high coating stability.2. DR-polystyrene sulfonate coating was prepared on the inner wall of capillary by replaced silane reagents with water-soluble diazo photopolymer resins (DR). Layer-by-layer self-assembly ultrathin films were formed through ionic bonding action, which ultimately converted into covalent bonds under UV irradiation, thereby forming a covalently bonded DR-PSS coating with self-assembled structure on the inner wall of capillary. The experimental results showed that covalently bonded coating has the capacity of inhibiting adsorption of protein.In this paper, the preparation techniques of coated capillary combine many advantages of physical adsorption and chemical bonding coating method. Covalently bonded coating with controllable structure can be formed by UV irradiation cross-linking. The results further proved that the preparation of coated capillary techniques is a type of procedure with the characteristic of simple and quick, efficient, reproducible and environmental protection.
