Matteuccia struthiopteris (L.) Todaro is an perennial herbaceous plant, belonging to family Onocleaceae and Pteris Matteuccia, which are existed widely. It is a kind of meicine edible plants, whose young leaves can be food and rhizome can be medicine. The fern contains flavonoids, polysaccharides, phenols, steroids, terpenoids, organic acids, proteins and other chemical composition, and polysaccharide is one of important composition. It is proved that polysaccharide in the rhizomes of the fern has immune biological activity.This study was designed to investigate the polysaccharide content, the ultrasonic extraction craft, monosaccharide composition, antioxidative activity, hepatoprotective effects on alcoholic liver injury in mice of polysaccharides extracted from the rhizome of Matteuccia struthiopteris (L.) Todaro (MSP). The primary results were as follows:(1) Using phenol-sulfuric acid method to determing the polysaccharide content, the methodological experiments showed that the method had high precision, stability, repeatability and recovery rate, and can be used as detection means of polysaccharide in Matteuccia struthiopteris. Comparing the extraction yield of polysaccharide in trophophyll, rhizome and sporophyl of different area, it is showed that the highest extraction yield of rough polysaccharides in trophophyll and rhizome were materals of Ning-shan and the highest extraction yield of rough polysaccharides in sporophyl was materal of Feng-yukou. Comparing the extraction yield of polysaccharide in trophophyll, rhizome and sporophyl of Ning-shan, it is showed that the distribution in different position was nonumiform and the highest extraction yield of rough polysaccharides was rhizome.(2) The influence of the factors such as ethanol liquid ratio, extraction temperature, ultrasonic time, ultrasonic power on the extraction rate of polysaccharide in rhizome of Matteuccia struthiopteris (MSP) were discussed. And the optimal process was confirmed by single and orthogonal experiments and listed as:the ethanol liquid ratio was1:50(g/mL), extraction temperature was80℃, ultrasonic time was30min, ultrasonic power was150W. In this condition, the extraction rate of the total flavonoids was9.61%. The influence of ethanol liquid ratio, extraction temperature, ultrasonic time, ultrasonic power on the extraction rate of polysaccharide in trophophyll of Matteuccia struthiopteris were significant difference at0.01level. The influence order of the factors was ethanol extraction temperature>liquid ratio>ultrasonic time>ultrasonic power.(3) Physical and chemical properties and monosaccharide components of polysaccharide was determined respectively. Monosaccharide components of MSP was analysised by HPLC. The results showed that the components of monosaccharide were glucose (Glc), galactose (Gal), and xylose (Xyl). Besides, the mole ratio between Glc:Gal:Xyl was3.502:1.392:1, and Glc was quantified to be in the highest level. MSP was pale yellow power, easy dissolve hote water and was polysaccharides by phenol-sulfuric acid method. IR apectra analysis showed that the polysaccharide have characteristic absorption peak.(4) Free radical-scavenging antioxidative activity of MSP was separately analyzed for DPPH radical, OH radical and reducing power. The result showed that the polysaccharide had high DPPH radical and OH radical scavenging activity and had high antioxidative activity, revealing a dose-effect relationship. The polysaccharide had high OH radical scavenging activity, revealing an IC50of0.74mg/mL. The polysaccharide had high DPPH radical scavenging activity, revealing an IC50of68.68μg/mL. The polysaccharide had high antioxidative activity at the range of0.1~0.5mg/mL.(5) The mice model were induced by perfusing intragastrically with50%ethanol. To study the effect of MSP by testing the concentration of Glutamate Pyruvate Transaminase (GPT), Glutaruic oxaloacetid Transaminase (GOT) and Triglyceride (TG)in serum, as well as the activity of superoxide dismutase (SOD) and the concentration of malondialdehyde (MDA) in liver were measured. The result showed that groups of different dosage has different degrees of effect on acute alcohol liver injury in mice.
