Matteuccia struthiopteris(L.)Todaro is a perennial herbaeeous plant,belonging to Onocleaceae MatteucciaTodaro,also known as legumes,fern rhizome,Cantonese cuisine,cucumber.The leaf has two type,the nutrion is 1?2 pinnate,the sporophyll volumes down and the shape looks like pod.It propagates species by sexual and asexual reproduction.The fern contains a variety of medicinal ingredients,such as polysaccharides,flavonoids,steroids etc.It is one of the main five cyrtomium rhizome.The all plant can be used as medicine,has insecticidal,hemostatic effect and so on.In addition,it is rich in protein,fat,carbohydrates,crude fiber and other nutrients.It is a kind of high nutritional value of green materials,which also collects medicinal and edible plants for the integration of resourse;In addition,the type is beautiful and elegant,has very high ornamental value,It is the excellent ornamental material in gardens and indoor.A large number of studies have shown that the triterpenes has many kinds effection,such as anticancer,antivirus and protecting liver.The triterpene is rich in fern,this paper studies the response surface which was used to optimize ultrasonic-assisted extraction of total triterpenes from Matteuccia struthiopteris by response surface methodology.And compared the difference of the total content of the triterpenoids of different parts,Separation and purification triterpenes from Matteuccia struthiopteris with AB-8 macroporous resin,orthogonal design was used to optimize this process.Through examining five kinds of anti-oxidation system,Analyzed and compared the total triterpenes antioxidant capacity,which were before and after purification;through the mouth give high concentrations of alcohol and increasing gradient method,set up two different type of acute and chronic alcoholic liver injury in mice respectively.The liver was injured,to detect the GPT,GOT,MDA and SOD levels through kit.Paraffin sections of mouse liver tissue lesions were observed,to provide theoretical basis in application of liver injury prevention and treatment,the results were as follows:(1)Optimization of ultrasonic-assisted extraction of total triterpenes from Matteuccia struthiopteris by response surface methodology,the optimal extraction conditions were determined as follows:ethanol concentration 60%,ratio of solid-liquid 1:35,ultrasonic temperature 63 ?,ultrasonic time 39 minutes.Respectively to analyze and compare the total triterpenoids content in three different parts of fern.The results show that:The total triterpenoid content in different parts that showed inhomogeneity,rhizomes>Nutrition leaf>spore leaf.(2)Separation and purification of triterpenes from Matteuccia struthiopteris with AB-8 macroporous resin,to optimize this process by orthogonal design.The best adsorption conditions parameters that optimizated are as fllows:the pH and concentration of sample was 6 and 1.96 mg/mL,the maximum injection volume was 5BV,the flow velocity was 2.0 mL/min,adsorption rate was 91.07%.The desorption process was determined as follows:60%ethanol was used as eluent at the flow rate of 2.0 mL/min with 4 BV.The recovery rate reached up to 86.27%,The purity increased from 28.61%to 82.32%,the purified multiples of triterpenes was 2.88.It showed that the adsorption and desorption rate of AB-8 resin is higher,purification was well.(3)Through comparing the antioxidant capacity,five kinds of antioxidant system experimental results showed that the order of antioxidant capacity was Vc>purified product>BHT>crude extract;Both total triterpenes and purificated triterpenes had stronger antioxidation effect on ABTS+,DPPH·,·OH.However,all of them had less effect on inhibiting O2-..It had a significant dose effect.The scavenging power of purificated triterpenes between BHT and Vc.(4)The GOT and GPT biochemical indicators illustrated purificated triterpenes had some extent inhibited and the inhibition was positively correlated with the dose,the alcoholic liver injury model was successful established.If some tissue or organ of body lesions occur will lead to some free radicals generated too much,cause serious damage to the body.The MDA and SOD biochemical indicators illustrated purificated triterpenes had strong antioxidant capacity in vivo.Its mechanism may be through enhanced liver antioxidant capacity of the body and effectively remove free radicals so as to achieve effectively alleviate the adverse damage to the liver caused by alcohol.Liver tissue paraffin sections showed that the medium and high doses purified triterpenoids can effectively relieve the liver lipid peroxidation,oxidative damage caused by free radicals attacks,reduced swelling,necrosis and fatty degeneration of the liver cells,regulating cells to restore normal metabolism and physiological functions,Its effect within a certain concentration range prefer than the positive control BP,The results showed the purified triterpenoids has a protective effect on acute alcoholic liver injury,the mechanism needs further study.
