Objective:To study the change of the endocrine function ofendothelial cell in Wistar rats with type2diabetes and the effects of aerobic exercisetraining on the endocrine malfunction of the endothelial cells and the possiblemechanisms.Method:45healthy male Wistar rats were8-week-old.13rats were selectedand randomly as normal glucose level group,which fed with normal standard diet.Therat model of type2diabetes mellitus was estabished by a single injection of low dosestreptozotocin(STZ,30mg/kg) via though abdominal after they were fed with highcalorie diet for7weeks in32remaining rats.The fasting plasma glucose,fasting seruminsulin and other corresponding indexes were measured to evaluate the quality of themodel.The normol glucose level group were randomly divided into2groups:normalcontrol group(C group) and aerobic exercise training group(CE group).The rats withtype2diabetes were randomly into2groups:DM control group(DM group) andDM+aerobic exercise train group(DME group).The rats in CE group and DME groupdid the exercise training which used60minutes’ unload swimming,6times per week.8weeks later,the indice of von Willebrand factor,plasminogen activator inhibitor-1,proteinkinase C and diacylglycerol were measured by enzyme linked immunosorbentassay.Result:1)After the establishment of type2diabetes model,compared with Cgroup,the FPG and HOMA-IR increased significantly,and FINS level decreasedmarkedly.Compared with C group,the plasma vWF and PAI-1concentration in DMgroup rised markedly,the aortic DAG and PKC also increased significantly.2)Aerobicexercise training could markedly reduce plasma vWF and Pai-1concentration.At thesame time,the aortic PKC and DAG concentration in DME group is significantly lowerthan the DM group.3)There are no significant difference between C group and CEgroup.Conclusion:1)The rat model of type2diabetes could be estabished by a singleinjection of low dose streptozotocin after they are fed with high calorie diet for7weeks.2)Aerobic exercise training could decrease the FPG and HOMA-IR in T2DM ratsas well as increase serum FINS level.3)8-week-swimming could inprove the endcorinefunction of endothelial cells by decreasing the synthesis of aortic PKC and DAG.
