Purpose: The purpose of this experiment is to study the relationship betweenthe expression of IER5in clinical cervical cancer tissue and the radiationdose,the affection of compound matrine injection on the expression of IER5incervical cancer cells to provide new theoretical basis for the research ofradiation sensitization effect and sensitization mechanism of Sophoraalkaloids.Method: The development of specimen collection program, a total of20casesof patients with cervical cancer tissue samples were collected from LiaoningProvincial Tumor Hospital gynecological oncology ward hospitalization fromSeptember2011to March2012, including the blank group,10Gy radiotherapy group,20Gy radiotherapy group,compound matrine injections combining10Gy radiotherapygroup, compound matrine injection combining20Gy radiotherapy group.There arefour cases each. RT-PCR was used to detect expressions of IER5mRNA. Theexpression of IER5in different groups was detected by immunohistochemistry.SPSS17.0statistical software was used to analyze the experimental data.One-wayANOVA was used to compare the differences between groups.Results:1. RT-PCR results showed the expression of IER5in the blank group、10Gy radiotherapy group、compound matrine injections combining10Gyradiotherapy group、20Gy radiotherapy group、compound matrine injectionscombining20Gy radiotherapy group was0.63±0.058、0.82±0.10、1.06±0.07、1.16±0.13、2.10±0.73. The level of the IER5mRNA in radiotherapy group is higherthan the blank group, the difference was statistically significant (P <0.05).Thelevel of the IER5mRNA in20Gy radiotherapy group was significantly higher than10Gy radiotherapy group,and the difference was statistically significant(P=0.001<0.05). The level of IER5mRNA in Compound matrine injection combined with radiotherapy group was significantly higher than the radiotherapy groupand the blank group,the difference was statistically significant (P <0.05). Andthe same radiation dose, the level of IER5mRNA in Compound matrine injectioncombined with radiotherapy group was significantly higher than the radiotherapygroup.2.Immunohistochemistry results showed the OD value of IER5in the blank group、10Gy radiotherapy group、compound matrine injections combining10Gyradiotherapy group、20Gy radiotherapy group、compound matrine injectionscombining20Gy radiotherapy group was0.210±0.015、0.223±0.005、0.243±0.003、0.250±0.003、0.271±0.010. The level of IER5in radiotherapy group is higherthan the blank group, the difference was statistically significant (P <0.05).Thelevel of the IER5in20Gy radiotherapy group was significantly higher than10Gyradiotherapy group,and the difference was statistically significant (P=0.001<0.05). The level of IER5in Compound matrine injection combined withradiotherapy group was significantly higher than the radiotherapy group and theblank group,the difference was statistically significant (P <0.05). And the sameradiation dose, the level of IER5in Compound matrine injection combined withradiotherapy group was significantly higher than the radiotherapy group.Conclusion:1. The condition of radiation raised the expression of IER5, andthere is a certain dose-dependent.2.Compound matrine injection combined with radiotherapy can upregulate theexpression of IER5, which may be related to the radiosensitivity of cervicalcancer organizations.
