Purpose:toobservetheeffectsofruanjianxiaoyinggraininautoimmunethyroidinflammation rats,through the level change of TgAb、TpoAb、apoptosis proteinsand the pathomorphology,to discuss the autoimmune thyroiditis pathogenesis.Material and method: inject Tg and IFA/CFA to immune and drink iodine waterto make animal mode,seprate rats with six groups,there were normal group、AITgroup、Tripterygium wilfordii Hook treatment group、the lower、secondary andhigher dose of ruanjianxiaoying treatment group.Except the normal group feedwater from beginning to the end.Other groups were given twice immunization bymixed Tg and complete Freund’s adjuvant one a week,in the same time to feedthem with0.064%NaI,the next four weeks,strengthen the immune ofthem,immunization by mixed Tg and incomplete Freund’s adjuvant,to make the AITmode.Give water the Tripterygium wilfordii Hook and the different dosage ofruanjianxiaoying grain for six weeks to the rats,detect the level ofautoantibodies TgAb and TpoAb with the electrochemical luminescence method、the level of apoptosis proteins(Fas、Fas-L、Bax、Bcl-2) by immunohistochemistrymethods、detect the apoptosis of grey value and observe the pathomorphology ofthyroid gland by light microscopy.Use the spss17.0to experiment the data ofstatistics.Results:1.detect the TgAb and TpoAbBefore the treatment,comparing every treatment groups and AIT group withthe normal group,TgAb and TpoAb were obviously elevated(P<0.01),comparingevery treatment groups with AIT group have no significant differences(P>0.05).After treatment,comparing every treatment group with AIT group,TgAb andTpoAb were obviously lower(P<0.01);comparing the secondary、higher dose treatment group and the Tripterygium wilfordii Hook treatment group with thelower dose treatment group have obviously differences (P<0.01);comparing thelower dose treatment group with the normal group have obviously differences (P<0.01);comparing the secondary、higher dose treatment group and theTripterygium wilfordii Hook treatment group with the normal group have nodifference(P>0.05);comparing the secondary、higher dose treatment group withthe Tripterygium wilfordii Hook treatment group have no obviously differences(P>0.05).2.observe the thyroid tissue under the light microscopyThe normal group:a large number of thyroid follicular and right size,thefollicular wall have the cuboidal epithelium to constitute double wall,theinterstitial have a little inflammatory cells,inside the cell have red jelly;The AIT group:comparing with the normal group, the follicular wall was thecircular,follicular volume was bigger,follicular wall was thin,boundaries arenot clear,in the center,the thyroid follicular volume was pyknotic,have a lotof inflammatory cells,inside the cell have differences color shades of red jelly;The Tripterygium wilfordii Hook treatment group:the thyroid follicular wasright size,follicular epithelium structure integrity and have double wall,havea little inflammatory cells,inside the cell have red jelly;The lower dose treatment group:the thyroid follicular epithelium structureintegrity,the follicular wall boundary was not clear,have a little inflammatorycells,inside the cell have red jelly;The secondary dose treatment group:the thyroid follicular have differencesize,the structure was normal,the follicular wall was full,epithelial cell linewas not clear,have a little inflammatory cells,inside the cell have red jelly;The higher dose treatment group:the thyroid follicular arrangement ofsparse,the follicular epithelial cells structural integrity and have double wall,have a little inflammatory cells,inside the cell have red jelly. 3.detect the apoptosis proteinsAfter treatment,comparing the AIT group with the normal group were obviouslydifferences (P<0.01);comparing every treatment group with AIT group haveobviously differences (P<0.01);comparing the secondary、higher dose treatmentgroup and the Tripterygium wilfordii Hook treatment group with the lower dosetreatment group have obviously differences (P<0.01);comparing the lower dosetreatment group with the normal group have obviously differences (P<0.01);comparing the secondary、higher dose treatment group and the Tripterygiumwilfordii Hook treatment group with the normal group have no difference(P>0.05);comparing the secondary、higher dose treatment group with the Tripterygiumwilfordii Hook treatment group have no obviously differences (P>0.05).4.the immunohistochemical of apoptosis proteins resultsThe apoptosis proteins of thyroid tissue were express inside the follicularepithelial cells,the cell membrane have a little express and the color was tan.The normal group:the thyroid follicular epithelial cells structuralintegrity,the size was right,distribution of a small amount dye;The AIT group:the thyroid follicular epithelial cells structural damage wereseriously,the size was not right,distribution of a lot amount dye;The Tripterygium wilfordii Hook treatment group:the thyroid follicularepithelial cells structural were less damage,distribution of a small amount dye;Every dose of ruanjianxiaoying group:the thyroid follicular epithelialcells structural were less damage,distribution of a small amount dye.Results:1.the TgAb and TpoAb of autoimmune thyroiditis were obviously elevated,thethyroid tissue were obviously damaged,explain the AIT mode were set up; Fas、Fas-L、Bax and Bcl-2have obviously differences,explain the phenomenon ofapoptosis were appear.2.Every treatment group could through reduced the TgAb and TpoAb to treatment autoimmune thyroiditis; the secondary、higher treatment group was close to theTripterygium wilfordii Hook treatment group;the secondary、higher treatmentgroup and the Tripterygium wilfordii Hook treatment group were better than thelower group;the secondary、higher treatment group and the Tripterygium wilfordiiHook treatment group were close to the normal group.3.Every treatment groups through regulation and control apoptosis proteins totreatment autoimmune thyroiditis.4.Every treatment groups could repair thyroid cell and reduce the pathologicaldamage.
