Purpose:Observed no dose of iodine Chinese herbal compound on experimental autoimmune thyroiditis (EAT) rat thyroglobulin antibody (TgAb), thyroid peroxidase antibodies (TpoAb), interleukin -1,4,6 and TNF-αand ultrastructure of thyroid tissue, thyroid pathology, thyroid regulation of apoptosis related proteins Fas / FasL, Bcl-2/Bax effects, screening out the best treatment of AIT dose of iodine-containing traditional Chinese medicine, and from the multi-target point to explore its mechanism.Material and method:Conventional 75 SD female rats, adaptive feeding week, 15 were randomly selected as control group, the remaining modules to build. In the model since the second week of drinking water iodine concentration of 500g / L, control group with distilled water. The fourth week of application modules built by the complete Freund's adjuvant (CFA) emulsified into a water in oil-like TG (TG: CFA = 1:1), a total of two subcutaneous in two-foot multi-point injection after an interval of two days, each time each 100μg, as the initial immunization, the fifth week of multi-point injected subcutaneously in the back by incomplete Freund's adjuvant (IFA) emulsion of TG (TG: IFA = 1:1), per each 100μg, for four weeks, as a strengthening immunity. Will become the model of the rats were randomly divided into model group (n = 13), Chinese medicine treatment of low dose group (14 rats), Chinese medicine treatment dose group (14 rats), Chinese medicine treatment of high-dose group (14 rats). Tang Xiaoying Ruanjian after 6 weeks of treatment, rats were sacrificed and abdominal aortic blood method to be adopted as the indicators measured: (1) enzyme-linked immunosorbent assay (ELISA) to detect the serum TgAb, TpoAb , IL-1, 6, TNF-αchanges; (2) was detected by immunohistochemistry in thyroid tissue from rats apoptosis regulatory proteins Fas / FasL, Bcl-2/Bax change; (3) were observed under light microscope morphological changes in thyroid pathology; (4) were observed under the electron microscope ultrastructural changes of thyroid tissue (5) using SPSS13 ? 0 statistical software to meet normality and homogeneity of variance between groups using analysis of variance, the data presented by x±s, the results of each group were statistically analyzed.Results:Chinese high, medium and low dose group and model group: Chinese medicine of serum thyroid autoantibodies TgAb, TpoAb and IL-1, 4,6, TNF-αlevels were significantly lower thyroid tissue Fas / FasL, Bax expression decreased , Bcl-2 expression in thyroid tissue increased. Light treatment group were observed structural damage of thyroid tissue in model group. Electron microscopy ultrastructural changes of thyroid treatment group in model group. The above data, the traditional Chinese medicine and high dose groups were superior to low dose effects, high, middle dose was no significant difference between groups.Conclusion:1.Different doses of Ruan jian Xiaoying has reduced serum levels of thyroid autoantibodies and the related role of cytokines.2.Different doses of Ruan jian Xiaoying thyroid gland in rats with ease EAT ultrastructural injury.3.Different doses of Ruanjian Xiaoying has to reduce the pathological injury in rat thyroid. 4. Different doses of Ruanjian Xiaoying has to reduce the role of rat thyroid cells.These results, the traditional Chinese medicine, the role of high-dose group were better than low-dose group, high and middle dose was no significant difference between groups.
