Hypo-function of ovary, also known as premature ovarian failure (POF), is the malfunction of the ovaries before age40in women, characterized by the sustained amenorrhea, the shrinking of sex organs, menopausal levels of follicle stimulating hormone (FSH), high luteinizing hormone (LH) levels and decreasing of estrogen (E2), in spite of that most of them have typical menstrual cycle, the age at menarche, slightly delayed puberty and normal development of secondary sexual characteristics.A commonly cited triad for the diagnosis is the perimenopausal syndrome symptoms:hot flushes, Daohan, facial flushing, emotional instability (including irritable or depression, low sexual desire, reducing of vaginal secretion, and dryness in vaginal), which will lead to infertility eventually.The clinical feature of POF belongs to the deficiency type of QI of the kidney."YI SHEN CU PAO TANG" is presented by Professor Niu Jianzhao in the Beijing University of Chinese Medicine, based on years of clinical experience. It has been widely used in clinic, and is very effective in treating the low of kidney qi deficiency type of ovarian function, even premature ovarian failure.OBJECTIVEEffect of YI SHEN CU PAO TANG on thelow of kidney qi deficiency type of ovarian function (POF) in model rats were studied by using animal experiments, and the mechanism behind the effect was also discussed.METHODSThe sexually matured SD rats with normal estrous cycle were chosen as the experimental objects. Firstly, these SD rats were intragastric administered for66days with Tripterygium Glycosides tablet60mg/kg/d, resulted in the malfunction of ovarian in rats, which can provide a useful model for studying the low of kidney qi deficiency type of ovarian function. The vaginal smear method was used to measure the estrous cycle, in order to identify whether these SD rats were qualified for the following study. Then, YI SHEN CU PAO TANG were used to treat these SD rats for2weeks. The general condition of rats (including the rat ovary, uterus, adrenal gland index, serum hormone FSH, LH, E2., P level, ovarian follicle, follicular, luteal size, luteal apoptosis protein Caspase-9and Apalf-1level) were measured and recorded.RESULTSAnimal experiments:(1) The uterus index, ovarian index and adrenal index in the qualified model SD rats was significant low when compared with that in the normal SD rats (P<0.05).(2) After the successful treatment, the number of corpus luteum in the model SD rats clearly decreased (P<0.05) when compared with the normal SD rats.(3) After the successful treatment, the circumference and areas of ovarian follicles in the model group were smaller than that in the normal group and clomiphene tested group (P<0.05).(4) After the successful treatment, the difference in blood serum FHS, LH and E2between the model group and normal group has statistical sense (P<0.05).(5) The difference in apoptosis protein caspase-9and Apaf-1between the model group and normal group has statistical sense (P<0.05).CONCLUSIONBy treatment using YI SHEN CU PAO TANG, the general feature of SD rats were improved significantly. The estrous cycle of model SD rats had been nearly normal again with weight increasing. Experimental results also show that the index of ovary and uterus increased, serum FSH and LH decreased, E2and P levels raised, apoptosis protein caspase-9and Apaf-1level reduced. All these results suggested that YI SHEN CU PAO TANG has a function similar to the estrogen and progestin, which can promote the recovery of ovarian function, enhance the growth of follicular and improve the overall health of reproductive function.
