Objective:To establish a quantitative method of multi-components by single marker(QAMS) for determining of Ginkgo biloba Extract and alkaloids of Jin ji Preparations. Selected a cheap, easy and stable composition as internal standard for the simultaneous determination of other ingredients, solve the problem that the other component reference material is not easy to get and to the durability of evaluation method is not good and difficult to apply to the actual production and test; improve and perfect the content determination method, as well as ensure the accuracy and feasibility of the method apply to medicinal herbs and traditional Chinese medicine preparation, and last accurately applied the method to practical production, testing and research.Methods:Choose10bacth of ginkgo biloba extract and35bacth of Jin ji preparations from five formulations, after pretreatment including μltrasonic extraction, then determine the content of seven flavonoids and three alkaloids with the typical composition quercetin and palmatine as the indexes by HPLC. Validate the accuracy of peak location using the relative retention value by liquid chromatography-mass spectrometry technology. Verified the accuracy of QAMS method using SPSS statistical software.Resμlts:The resμlts showed that the content of ginkgo biloba extract (mg/g) by QAMS:rutin content are30.60-39.38, quercetin content are1.22-2.14, luteolin content are0.05-0.12, kaempfeol content are0.44-1.14, apigenin content are0.11-0.20, isorhamnetin content are0.33-0.68;The content of ginkgo biloba extract (mg/g) by External standard method: rutin content are30.90-40.54, quercetin content are1.25-2.20, luteolin content are0.05-0.12, kaempfeol content are0.44-1.14, apigenin content are0.11-0.21, isorhamnetin content are0.33-0.67.After analyzing by liquid chromatography-mass spectrometry mass spectrometry, a mass spectrometry of the test component peaks and reference substance peaks are same.The Pearson correlation coefficient of rutin, quercetin, luteolin, kaempfeol, apigenin, isorhamnetin respectively are0.998,0.999,0.993,1.000,0.992and0.999, calcμlated value and estimated value did not make significant difference on QAMS and external standard method.The alkaloids content of Jin ji capsμle (μg/pill) by QAMS:inner reference palmatine content are35.15-94.18, jateorhizine content are54.85-117.55, berberine content are24.27-70.08; External standard method in Jin ji capsμle:jateorhizine content are55.03-119.0l,berberine content are23.90-64.89; The Pearson correlation coefficient of jateorhizine and berberine were1.000and0.998, and calcμlated value and estimated value did not make significant difference on QAMS and external standard method.Investigated the durability of the relative correction factor, the relative correction factor of the determination of flavonoids ingredients from ginkgo biloba extract and alkaloids ingredients from Jin ji preparations is accurate、effective、authentic.Conclusion:There is no significant difference between QAMS and external standard method. It is show that the relative correction factor is credible. In the case of lacking reference substance, we coμld detect the quantitation of Quercetin, then take advantage of the RCF to quantitate seven flavonoids; also detect the quantitation of palmatine, then take advantage of the RCF to other alkaloids,and this is feasible.Improved the reproducibility of relative correction factor by wavelength scanning,it helpfμl to application research of traditional Chinese medicine (TCM) and their preparations, and lay the foundation for the QAMS method extending to simμltaneous determination of different structural components.
