Recently Ginkgo leaves are having been one of the most popular traditional Chinese medicines (phytomedicines). In this paper, different analytical methods, such as capillary zone electrophoresis (CZE),high performance liquid chromatography (HPLC), liquid chromatography-mass spectrometry (LC-MS) and capillary electrophoresis-mass spectrometry (CE-MS), were set up to analyze the Ginkgo leaves and their extracts. The fingerprints of extract of Gingko leaves by CZE^ HPLC arid HPLC-MS methods were developed respectively.1 The determination of two flavonol glycosides in Ginkgo leavesThe composition of running buffer in CE method was optimized by using the uniform design U6 (36), and the influence of temperature on separation was also studied. The final separation condition was 500 mmol L-1 H3B03-50 mmol L-1 Tris-15 % (v/v) methanol (pH 8. 5). 30.0 kV and 25 癈 were applied. The linear range of rutin was 0. 022?. 353 mg ?ml"1 (7=0. 9994) and the average recovery was 98. 3 %. It was found by CE method the contents of rutin and isoquercitrin in the leaves of small Ginkgo trees in April were higher than that in others. In this paper, HPLC method was also used to determine the two flavonoids. The separation was carried out on a Zorbax SB C18 column using a gradient elution program with 0.5 % formic acid and acetonitrile. The results of these two methods didn' t show obvious difference by t-test.2 The analysis of active components in Ginkgo leaves by LC-MS/MS and CE-MS/MSThe main two kinds of active compounds in Ginkgo leaves wereAbstractanalyzed by LC-MS/MS simultaneously. Most flavonoids and terpene lactones were identified or deduced by comparing the retention time, UV spectra and MS spectra with that of standards. The library of 2nd MS spectra of the standard extract of Ginkgo leaves (EGb) 761 was set up, which is helpful to identify the compounds in fingerprint analysis.The method of CE-MS/MS was used on EGb analysis for the first time. The buffer and sheath liquid containing volatile electrolytes were optimized to get better ionization efficiency. Seven flavonoid compounds were detected. 3 The study on fingerprint chromatogram of EGb samplesIn this paper, the CE method and LC-MS method was used for the first time to study the fingerprint of EGbs samples. Three series of fingerprint chromatogram were set up by use of the CE-UV, LOUV and LC-MS method separately. The similarity of different EGb samples in contrast with EGb 761 was calculated by "fingerprint chromatogram similarity software" with rutin as the internal reference peak. It is thought that the samples whose similarity were below 0. 8 differed from EGb 761 obviously. The similarity results by three methods were contrasted and discussed.
