Objective:Explore medicated serum of kong JianGu pieces are in vitro culture of osteoblast proliferation and differentiation,To provide experimental basis for traditional Chinese medicine in the treatment of osteoporosis.Methods:Chinese medicine serum pharmacology method preparation of king kong JianGu piece of traditional Chinese medicine medicated serum in rats; Using multiple enzyme digestion method in vitro of osteoblasts; The four groups medicated serum joined with the newborn rat osteoblast culture respectively, to study their effects on osteoblast proliferation and differentiation and mature;Results:(1) the osteoblast morphological observation showed that24h visible cells were fusiform or polygon; Into osseous induced after5d, obviously increase the cell number was observed and volume began to increase, and proliferation, confluent;(2) Under the electron microscope observation of osteoblast alkaline phosphatase staining positive rate, visible experiment each drug group than the control group (normal saline group) staining obviously; Comparison between the experimental group, king kong JianGu group and ray nai acid strontium positive rate is most significant, no obvious difference between the two;(3) King kong JianGu group and ray nai acid strontium of mineralized nodule number and nodule area significantly better than the control group and loose bone treasure group, and no obvious difference between the two; (4) Experimental drug groups each fluorescence microscope:osteoblasts in type I collagen staining were expressed positive, quantity of type I collagen and intensity has obvious difference with the control group. Each drug group in king kong JianGu piece and ray nai acid strontium group dyeing is the most significant;(5) Cell culture1d,2d,3d detection. Cultivation of1d test, the experiment each drug group and control group in cell proliferation rate was statistically difference (P<0.05), with the western medicine control group ray nai acid strontium group was most pronounced; Cell culture,2d and3d detection, the experimental group and control group significantly (P<0.05), the difference from the previous king kong is compared between group JianGu pieces of bone cell increment rate than ray nai acid strontium, loose bone treasure group significant difference (P <0.05);Conclusion:(1) this study obtained cells capable of osteoblast, osteogenesis functional protein expression, in line with the typical features of osteoblasts, can be used in the subsequent experiments;(2) King kong JianGu can promote osteoblast maturation, differentiation, its effect and no obvious difference was found between ray nai acid strontium, but stronger than loose bone treasure;(3) Just JianGu can promote the proliferation of osteoblast, starting with the most obvious, ray nai acid strontium kong JianGu times,As the training time, king kong JianGu piece for the promotion of osteoblast, enhanced its positive effect is better than ray nai acid strontium and loose bone treasure。...
