Objectives:This experiment studies on traditional Chinese medicine Jianpi Xiaoai Fang’s effect on nude mice colorectal liver metastasis model VEGF and VEGFR-2factor expression. Primary discussion for its mechanism of action is progressed. It provides a foundation for new traditional Chinese medicine for prevention and treatment of tumor metastasis and relapse with high efficiency and low toxicity.Method:(1)use orthotopic HCT116cell injection, between the mucosa and the muscularis layers of the cecal wall of nude mice, make the model of livermetastasis of nude mice colorectal cancer.(2)nude mice are randomly allocated to group of Jianpi Xiaoai Fang, positive control, models and blank control; observe each group’s general situation, weight, survival and situation of liver metastasis; determine the expression of nude mice VEGF and VEGFR-2protein;(3) count and analyze experimental data, analyze Jianpi Xiaoai Fang’s influence on the expression of VEGF and VEGFR-2.Results:(1)Cecum submucosal injection of human colon cancer cell lines HCT116cells to establish nude mice model of colorectal liver metastases has a extremely high success rate, pathological histology confirmed the lier cancer cells presented feature of poorly differentiated adenocarcinoma;(2) The group of Jianpi Xiaoai Fang nude mice survival significantly longer than group of Positive control and models, the difference is statistically significant;(3)survival of the group of Jianpi Xiaoai Fang is obviously longer than the same of the groups of positive control and models,the difference has statistically significant;(4) The group of Positive control and models each have had10nude mice liver metastases, The group of jianpixiaoaifang have8nude mice liver metastases;(5) the expressions of VEGF and VEGFR-2protein are significantly lower than the groups of positive control and models, the difference has statistically significant.Conclusion:Jianpi Xiaoai Fang can significantly improve quality of life of liver metastasis model nude mice following the operation of colorectal cancer and prolong their survival period. It has a tendency to inhibit liver metastasis for experimental models. Their functions may be related to the down-regulation of VEGF and VEGFR protein.
