Psoriasis Classic Recipe Containing Serum Interleukin-22 Induced HaCaT Cell Proliferation

Psoriasis (PS) i s a common chronic, inflammatory, recurrent derma tosis which occurs2%-3%population of the world.PS is characterized by erythema and scales,brings serious impact to the patients’ physical and mental health by attacking young adults mostly.The etiology of PS is not clear yet, the security and long-term efficacy of western medicine is not ideal,while the traditional Chinese medicine (TCM), with less side effect, lower recurrence rate, increasingly welcomed by the majority of patients and clinical medical. The domestic reports of TCM therapy for PS in clinical are very rich, while reports for mechanism of the classic prescriptions are few.This paper used interleukin-22(IL-22) and classic prescriptions as starting point, explored immune pathogenesis in PS and the mechanism of classical prescriptions for PS, mainly included two parts: literature reviews and experimental research.There were two literature reviews. The first one reviewd the progress of the relationship between IL-22and PS, includes source, gene, protein structure, sign al pathway of IL-22, summarized evidence, pathogenesis and application of IL-22in PS; The second one was about serum pharmacology of TCM, reviewd the preparation and application of serum pharmacology,as well as its advantages and deficien cies.ObjectiveTo explore whether IL-22can promote the proliferation of KC and its related molecular mechanism,to clarify the role of IL-22on the pathogenesis mechanism of PS; To observe pharmacological serum of classic TCM prescriptions (Xijiaodihu ang Decoction, Taohongsiwu Decoct ion, Dangguiyinzi Decoction) on HaCaT cells (hum an immortalized keratinocytes)induced by IL-22,to explore the mechanism and target of classic TCM prescriptions treating PS,providing theory basis of classic TCM prescriptions treating PS yulgaris in clinic.Methods1Preparation of classic TCM pharmacological serums, including Xijiaodihuang Decoction group, Taohongsiwu Decoction group, Dangguiyinzi Decoction group, MTX group and control group.2Detect effective concentration of IL-22by the rate of HaCaT cell proliferation using MTT method.3Survey activated molecular pathways of IL-22on HaCaT cells by Western Blot method after adding effective concentration of IL-22.4Observe the influence of classical TCM pharmacological serums on IL-22induced HaCaT cells by MTT method after adding effective concentration of IL-22 and5different classic TCM pharmacological serums.Results112.5-200ng/ml IL-22can stimulate proliferation of HaCaT cells. Compared with control group,12.5ng/mL,25ng/mL IL-22groups showed no statistical differences on the proliferation of HaCaT cells (P>0.05), while the rest groups had significant differences(P<0.05). When the concentration of IL-22was below150ng/mL, its effect on promoting HaCaT cell proliferation has positive correla tion with concentration of IL-22; while above150ng/mL, it turned out to be opposite way.2with50ng/mL IL-22which was the effective concentration treating HaCaT cells in O min to120min, we can see the phosphorylat ion of JAK1and STAT3pathway proteins, pathways open in30min, largest in60min, began to attenuate after120min.3. Xijiaodihuang Decoction group, Taohongsiwu Decoction group, Dangguiyin zi Decoction group can inhibit the proliferation of IL-22induced HaCaT cell model. The rate of inhibition increases with higher concentration of serum, showe d a positive correlation; The rate of innibition was different while culture time changes,and48h culture had strongest effect; the impacts of inhibition were different among4pharmacological serums at the same time and concentration,they were Xijiaodihuang Decoction>Taohongsiwu Decoct ion>MTX>Dangguiyinzi Decoction,3classic TCM serum groups had no statistical differences with MTX group(P>0.05).Conclusion1IL-22can promote proliferation of HaCaT cells in dose dependent way by activat ing JAK1/STAT3molecular pathways, IL-22was used to imitate the condition of KC’s disorder in PS and showed the model stability.2classic TCM can inhibit the proliferation of IL-22induced HaCaT cells model, revealed the theory basis of classical TCM treating PS from inhibiting hyperplasia of KC. Prompted rationality of "based on blood" in the diagnosis and treatment of PS.
