Objective:This research was aimed at studying the effect of cluster needling of scalp acupuncture on the neurological function score and the functional protein expression of rats with right cerebral ischemia after MCAO model, and therefore exploring the molecular mechanism of cluster scalp acupuncture on post-stroke neuronal plasticity by protein level.Methods:The experiment selected36healthy inbred male SD rats, weight between240-260g, skin smooth and moist and airway open. All the rats were provided by Department of Experiment Animals, Capital Medical University. With random numbers, the experiment divided into the following groups:1d sham operation group,1d model group,1d cluster needling group,14d sham operation group,14d model group and14d needling group. The focal ischemia rats models were made by middle cerebral artery occlusion (MCAO) with monofilament (improved Zea-longa Method). The rats in cluster needling group were treated with cluster scalp acupuncture everyday on the parietal area for30min per time. No supplement if any rats in the above six groups died. Implement the Bederson Evaluation on the24nd hour and the14th day post the operation. Collect the brains after evaluation and find the expression changes of functional proteins in cerebral ischemia rats by Western-blot method.Results:1. Behavioral score:on the24nd hour, the differences among the model group, the cluster scalp acupuncture group and the sham operation group were statistically significant (P<0.05), indicating the models were successfully made; the difference between the model group and cluster scalp acupuncture group was not statistically significant (P>0.05), showing comparability; on the14th day, the behavioral score of cluster scalp acupuncture group was superior to the model group, with difference statistically significant (P<0.05), indicating cluster scalp acupuncture can promote the motor function recovery in rats. 2. GFAP protein expression:on the24nd hour, the differences among the sham operation group, the model group and the cluster needling group was not statistically significant (P>0.05). However, the GFAP protein expression means of both the model group and the cluster needling group were higher than the sham operation group; on the14nd day, the difference between the cluster scalp acupuncture and the model group was not statistically significant (P>0.05). The GFAP protein expression mean of the cluster needling group was lower than the model group, indicating cluster needling tends to decrease the GFAP protein expression.Conclusion:It can promote the motor function recovery in rats and tends to decrease protein expression by cluster needling of scalp acupuncture.
