Objective:Research the effects of downhill run in growth period on blood index, Bone mineral density (BMD), collagen I, Bone marrow Mesenchymal stem cells (MSCs) and induced the Osteoblast(OB) differentiation in female ovarian older mice. Simulating the postmenopausal women by ovariectomized mice,use the technologies of RT-PCR, immunohistochemical and cells cultivate, systemly research the downhill run in growth period on ovariectomized mice bone from the biochemical index, bone mass, protein and cellular level. Search an effective sports model and provide theoretical support for erercise in growth period to prevent postmenopausal women osteoporosis happening.This study will also provide theoretical for the effects of training in growth still exist after a period of time.Research methods:After one week rehabilitating, dividing805-week old female mice into five groups:Control group (group C, n=16), Downhill group (D group, n=16), Control to ovarian group (C+OVX group, n=16), Control to SHAM group (C+SHAM, n=16) and Downhill to ovarian group (D+OVX group, n=16). The D gro-up and D+OVX group do downhill run movement in the early stage for eight weeks (0.8Km/h,40min,5d/week). After eight weeks, sacrificed the mice of the D and C groups and test experiment indexes. After the eighth week, C+OVX group and D+OVX group mice do ovarian surgery, C+SHAM group do SHAM Surgery. Postoperative sanatorium six weeks, after rehabilitating executed mice and test the same index test. Testing index includes:weighing the mice weight and food intake on fixed time each week; Using kit and the enzyme sign meter to test alkaline phosphatase (ALP) concentration, calcium and phosphate in blood; Using Hol-ogic Discovery A bone mineral density meter to scan the BMD of pectoral limb skele-ton; Using the technology of RT-PCR to detecte the expression of COL1gene in the bone; Using immunohistochemical method and technical of the paraffin section to inspecte the expression of collagen of the bone; cultivating the mesenchymal stem cells from marrow of tibial and femoral and inducing OB differentiation, using ALP dyeing to test MSCs quantity and Von Kossa dyeing to test OB osteogenesis ability.Result:Compared with the group C, the mice of D group have higher weight and food intake, higher serum ALP concentration (P<0.01), lower blood calcium concentration (P<0.05), higher blood phosphorus concentration (P<0.05), higher bone mineral density (P<0.05), higher bone tissue COL1expression in quantity (P<0.01), more bone collagen protein content, obviously the number of MSCs is larger and the area OB dyeing is large too, there are significant differences. This suggests that the downhill run exercise has good stimulitive effect on BMD, MSCs and osteo-blast. Compared with C+OVX group, the mice of D+OVX group,s weight and food intake does not exist significant differences, serum ALP concentration is higher (P<0.01), have lower blood calcium concentration (P<0.05), higherer blood phosphorus concentration (P<0.05), higher bone mineral density (P<0.05), bone tissue COL1expression is obviously higher in quantity (P<0.05), have more bone collagen protein content, obviously the number of MSCs and the area of OB dyeing is larger, there are signifycant differences. This suggests that the downhill run movement on the growth and ovarian mice have good promote role,including BMD, MSCs and OB. But most of the indexes after the surgery all appear largely change, are bad for bone health, may be its the effect of ovarian induce estrogen lack. Compared with SS group, D+OVX group and C+OVX group all indexes such as BMD are exist significant diff-erences. D+OVX group D group, C+OVX group compared with S group more, the indexes such as BMD are also exist very significant differences. This suggests that bone absorption more quickly than bone growth after ovarian, in some extent, move-ment can make up the influence of lack estrogen after ovarian on bone.Conclusion:1. Downhill run training can obviously stop weight increase of growth mice, and also make food intake increases; ovarian can cause weight rapidly increase in mice.2.Downhill run training can increase the minerals absorption of bone, make serum calcium and phosphorus concentration decreased, the level of serum alkaline phosphatase increase; ovarian can make serum calcium and phosphorus concentration increases, the concentration of alkaline phosphatase decline.3. Running can improve the bone synthesis metabolism of mice, so increase the bone mineral density; Ovarian make bone decomposition metabolic rate greatly increase and significantly decline the bone mineral density.4. Training in growth can increase the quantity of bone marrow stromal stem cells in mice, and increases the osteogenesis ability of osteoblast; Ovarian can reduce mesenchymal stem cells in the bone marrow and the osteogenesis ability of osteoblasts.5.Downhill run can make COL1mRNA up-regulate and increase synthesis of I type collagen in osteoblasts; Ovarian can reduce COL1mRNA expression and the reduce the synthesis of I type collagen in osteoblas.6. The I type collagen protein in bone tissue can increase mineral deposits in bone, increase bone mineral density and improve peak bone mass.7. The downhill run sports is a kind of effective mode to bone health, can effectively increase the peak bone mass value of teenagers, have important meaning to prevent osteoporosis happen.
