| This study was designed to explore the role and mechanism of early exercise intervention on bone metabolism in ovariectomized mice, and analyse the impacts of early exercise on bone metabolism. Ovariectomized mice by surgery is used to simulate postmenopausal women’s bone mineral status, and we can analyse the impacts of early exercise intervention on bone mineral density bone, bone structure, metabolic markers and mRNA expression of osteoblast-related factor in ovariectomized mice, and explore the reson,in order to provide a reference for the prevention and treatment of osteoporosis in postmenopausal women indirectly.[Objective]1. Establish the downhill running exercise model,in order to provide the reference for exercise and bone health related research.2. By ovariectomized mice model to simulate postmenopausal osteoporosis, to study the movement on the impact of osteoporosis in postmenopausal women.3. Study on the early downhill running exercise on bone mineral density, bone structure, bone metabolic markers and osteoblast-related factors (TGF-β、Smad3、 OC) gene expression in ovariectomized mice.[Method]Close40C57BL/6female mice about5-weeks-old for the study randomly and divide into5groups:control group, sport group, sham group,control+ovx group, sport+ovx group, n=8. After8weeks training of sport group and sport+ovx group, the mice of control group and sport group were slaughtered and tested bone mineral density (Kodak dual energy X-ray absorptiometry), bone structure (Processing vertebral samples for Methylmethacrylate embedding), bone metabolic markers (721Spectrophotometer), and osteoblast-related factor mRNA expression (RT-PCR). The mice of sham group were sham treatment, control+ovx group and sport+ovx group were ovariectomized. After6weeks rest quietly of this three groups were slaughtered and tested index, the same with the first time.[Result] 5. Early exercise intervention on bone health in ovariectomized mice of old age have certain effect. |
