Objective:To observe the histopathology change and IL-1β、PGE2level of Spragu-Dawley rat by using Medical hospital late well-known weapons experts Professor Zheng Huaixian’s ZhengShiYiHaoXunXiYao in treatment of acute soft tissue injury. And to study the pharmacodynamic action and mechanisms of ZhengShiYiHaoXunXiYao, Providing a theoretical basis for clinical treatment.Methods:48Kunming mice were divided into four groups at random, which were blank control group, ZhengShiYiHaoXunXiYao high dosage group, middle dosage group and low dosage group, observing hot-plate method to the ache of mice with Anti-bruise powder for comparison.48Kunming mice were divided into four groups at random, which were blank control group, ZhengShiYiHaoXunXiYao high dosage group, middle dosage group and low dosage group, observing analgesic effects by using tacetic acid writhing test.72SD rats, weighing220±20g, model by fight method after laboratory adaptive feeding7days. Using Random number table to divide into two groups (n=36), the experimental group (ZhengShiYiHaoXunXiYao) and the blank control group. Giving the experimental group with ZhengShiYiHaoXunXiYao treatment twice a day when modeling and the blank control group with water fumigation twice a day after24hours. Every12rats are killed on the1th、3th、5th day after using dosage and establishing rats acute soft tissue injury model and randomly selected, to observe the index change of the soft tissue injury area and to observe the changes in tissue morphology by skeletal muscle embedded in paraffin using light microscope. To measure the level of IL-1β and PGE2in. tissue homogenate by ELISA function. To observe the lesion of the muscle tissue using MR-DTI scan on the1th、3th、5th day after using dosage and establishing.Results:Every ZhengShiYiHaoXunXiYao dosage group could increase the pain threshold of the mouse hot plate method, and even has the significant difference with the model control group. The high dosage drag group and the middle dosage group significantly decreased the number of twisting body in mice with acetic acid affecting. There are high significant difference between the high dosage group and the model control group. The injury symptom index indicated every ZhengShiYiHaoXunXiYao group increasing the pain threshold of the mouse hot plate method, the significant difference with the model control group. ZhengShiYiHaoXunXiYao could promote the dissipation of local swelling damage, ecchymosis and subcutaneous hemorrhage, and has significant difference with blank control group. NMR Photo shows the ZhengShiYiHaoXunXiYao significantly and quickly reducing acute soft tissue injury inflammation and edema compared with the model control group. Observed under light microscope, repair of muscle fiber has quickly happened by using the drag. The drag could significantly inhibit the increasing of injury soft tissue PGE2and IL-1β on the1th、3th、5th day and high significant difference compared with model group.Conclusion:ZhengShiYiHaoXunXiYao has the function of analgesic and detumescence improvement and necroses evidently decreased. The probably mechanism in treatment of acute soft tissue injury is what ZhengShiYiHaoXunXiYao can reduce the concentration of IL-1βand PGE2in injured part, probably by this way achieving the action of analgesic and detumescence. It is worth of promotion and application in clinical.
