Malus prunifolia(wild)Bovkh(Qiuzi in Chinese,Malus)grows in some Chinese areas such as Hequ Pianguan of Shanxi Province,Baode and Fugu of Shannxi Province.The fruits of Malus prunifolia(Wild)Bovkh have been used as Fructus Crataegi in some area with good benefit for the blood liquids of the old.Some health care product is made from its fruit The current research on composition of Malus prunifolia(wild)Bovkh.There have no reporter of the flavonoid in Malus prunifolia(wild)Bovkh most on the initial description of nutrients. But deep study especially research was not reported in the literature on the flavonoid compounds.In this experiment,we use the Malus prunifolia(wild)Bovkh from Hequ of Shanxi as raw material, In this paper, on the basis of single factor test, Malus prunifolia(wild)Bovkh as raw material and orthogonal experimental design was determined about Malus prunifolia(wild)Bovkh of total flavonoids,and flavonoids compounds extraction process was studied.Using the polymide column chromatography thchnology to separate and purity the flavonoids,and doing the antioxidant test.In order to provides certain theoretical foundation for Malus prunifolia(wild)Bovkh development and utilization.The result of experiment as follows:(1) Taking ethanol as solvent to extract flavonoids from Malus prunifolia(wild)Bovkh is studies.During the reflux extraction process,four main factors including extracting time,extraction temperature,ratio of solvent to sample and extraction solvent concentration were stuied.By using single factor ananlysis method and the orthogonal analysis,The most influential factors is the concentration odethanol and far more then the other3factors.The effect of extraction time and extraction temperature on extraction is very close to the flavonoids of Malus prunifolia(wild) Bovkh.The minimal effect of extract flavonoids of Malus prunifolia(wild) Bovkh is ratio of material to liquid.The optimal condition of solvent extraction were obtained as follows:ethanol concentration80%、the extracting3h、ratio of material to liquidl:30、 the extraction temperure40℃, extracting for three times and the extraction rate is5.27%. (2) From using TCL chromatography,founding the most developing agent is Butanol:ethyl acetate:water(4:1:5);Preliminary judgment flavonoids of Malus prunifolia(wild) Bovkh probably contain melim.The extracts of the Malus prunifolia (wild) Bovkh flavonoid were rudely separated by different organic solvent. We can obtain the extracts of ethyl acetate,the extracts of trichloromethane,the extracts of butyl alcohol and the extract of waterphase.Then the extracts of ethyl acetate were again separated by Polyamide column chromatography.We obtained several flavonoids components,which were examined by High Performance Liquid Chromatography.Three flavonoids(B, C, F) components material purity reached more than90%.Then we can identify their structures and infer the B maybe melim, C maybe quercetin. Because the material F is very small quantities.NMR analysis failed to analyze results.(3) The antioxidant activies of flavonoids from Malus prunifolia (wild) Bovkh was researched by.OH、O2.-、DPPH’.The result showed that the extracts of ethyl acetate,the extract of trichloromethane,the extrects of butyl alcohol and the extracts of ethanol had stronger ability to scavenge radical,the effect are weaker than Vc.The sequence of scavenging efficiency were as follow:The total reducing capacity:Butanol<ethyl acetate<chloroform<the aqueous residue<Vc.In the system of02.-:chloroform<ethyl acetate<Butanol<the aqueous residue<Vc.In the system of OH:chloroform<ethyl acetate<Butanol<the aqueous residue<Vc.In the system of DPPH.:Butanol<ethyl acetate<chloroform<the aqueous residue<Vc.In conclusion,flavonoids from Malus prunifolia (wild) Bovkh have strong antioxidant activity.
