Biological significance of mercury ions and small molecules thiols playsimportant physiologicals role in life; the specific detection of these components hasimportant practical significance in interpreting its biological function. Among variouskinds of test, the fluorescence probe method shows important application value in thestudy of biomedical and life sciences because of its high sensitivity, high selectivity,fast and convenient and can be combined with fluorescence microscope imaging ofcells and living in situ determination of physiological activity components inbiological samples in real time, etc. In this work, several novel Fluorescent Probeswere designd and synthesised by modifying the fluorophore with good photophysicalproperties.A kind of mercury (Hg2+) ions fluorescent probe YG-1was designed andsynthesized with simple synthetic steps and high selectivity and sensitivity to mercuryions. After the probe combined with mercury ion, fluorescence emission spectrumranges from547nm to534nm, blue shift of13nm. The probe emitted a strong greenfluorescence, fluorescence of its solution changes from yellow to green. Using theprobe, it is successful to detected the mercury ion selectively in ratio in the watersoluble organic (CH3CN/HEPES=1:1, pH7.4).A colorimetric and turn-on selectively detecting of thiols (Cys and the Hcy, andGSH) fluorescent probes SY-1was designed and synthesized. The probe wassynthesized just three steps, starting from4-naphthalene-1,8-anhydride. After theprobe reacted to copper ions, color of solution ranges from colorless to light yellow,the absorption peak shifts from418nm to the490nm, clear red shift (72nm), andfluorescence quenching occurs at541nm. The probe using for the detection of smallmolecules thiols possesses good selectivity, high sensitivity, response speed, andother advantages. It has been successfully used in fluorescence imaging of CHO cells(Chinese Hamster Ovary Cells). The result shows that SY-1is an effective fluorescent probes detecting small molecule thiols in the cell.
