This paper reviews biological activity and extraction technology of phlorotannins, research the general situation of Sargassum fusiforme.This paper taked sargassum fusiforme as experiment materials, the optimization of extraction technology of polyphenols from Sargassum fusiforme by response surface method,separation and purification of polyphenols by macroporous adsorption resin, preliminary study on Sargassum polyphenols antioxidant activity in vitro,and liquid chromatography mass spectrometry(LC-MS) to analyze the main components of Sargassum fusiforme polyphenols. The main conclusions are as follows:In order to extraction technology optimization of sargassum fusiforme polyphenols,investigation of single factor extraction liquid volume fraction, extraction temperature, extraction time,solid-liquid ratio, extraction times on extraction rate of sargassum fusiforme polyphenols.Using the response surface analysis method of 3 factors 3 levels, and the leaching rate as index,the optimum extraction conditions for: ethanol concentration of 40%, extraction time of46 minutes, ratio of liquid to material 20:1, temperature of 70℃, under these conditions,sargassum fusiforme polyphenols leaching rate is 4.048%, and the theoretical value 4.050%,the error between them was 0.04%, verified the model can better predict the actual leaching rate.The results show that the static adsorption and desorption experiments were screening on 10 different types of macroporous resins: XDA-7 macroporous resin showed the adsorption and desorption capacity of the best. The experimental results show that when the sargassum fusiforme polyphenols column chromatography concentration of sample was 3.2mg/mL, sample flow rate was 1BV /h, and ethanol solution was 60%, flow velocity was 2BV /h, elution volume was 5BV, obtained sargassum fusiforme polyphenols the purified sample was 61.86%.Antioxidation experiment in vitro of purified sargassum fusiforme polyphenols, the results showed DPPH radicals scavenging capability of Sargassum fusiforme polyphenols IC50 was 6.65μg/mL; inhibition of superoxide anion radicals IC50 is 6000μg/mL; hydroxyl radical inhibition by IC50 was 125.42 μg/mL; the total antioxidant capacity was 65.04U/mg. On the whole, sargassum fusiforme polyphenols have strong antioxidant activity.Using liquid chromatography mass spectrometry(LC-MS) analysis of the main components of Sargassum fusiforme polyphenols, The experimental results showed that Sargassum fusiforme polyphenols has 7 main components, combined with the [M+H]+ molecular level map using Xcalibar software comes with the fitting software, the molecular formula and molecular structure,mainly containing hydroxyl and phenolic compounds.
