The great progress has been made in research on pharmacological activity of Sargassum fusiforme in recent years.At present,the domestic and foreign research on Sargassum fusiforme mainly focused on its anti-tumor activity and anti-coagulation activity,However,there are few reports about its moisturizing activity and its application in cosmetics.Sargassum fusiforme is rich in resources,and fucoidan from Sargassum fusiforme(FSF)has high research value in daily chemical industry.In this paper,Sargassum fusiforme was used as raw material to extract fucoidan by cellulase hydrolysis-ultrasonic assisted extraction method.after further separation,purified polysaccharide fragments was received,then evaluate its moisturizing effect.The main research contents are as follows:(1)FSF was extracted by cellulase hydrolysis-ultrasonic assisted method.Single factor and orthogonal experiments were used to optimize the extraction conditions.The optimum extraction conditions were as followings: pH,4.5;enzyme dosage,1%;extraction temper-ature,40 ?;cellulose hydrolysis time,2 h;ultrasonic time,15 min;and material-liquid ratio,1:10.Under the optimal condition,the extraction yield of FSF was 8.50%,and the yield of crude extract of fucoidan from Sargassum fusiforme(CEFSF)was 15.48%,The confirmatory experimental results show that the process is reproducible.(2)CEFSF was separated and purified by DEAE-cellulose and Sephadex G-150,then FSF-1 and FSF-2 were achieved,and the yield of them were 0.938% and 1.4817%,respect-tively.The structure of FSF-2 was analyzed,the results showed that: its Mw was 103418 Da,it was a kind of pyranose polysaccharides with ?–type glycosidic bond,the content of sulphate radical and uronic acid were 3.99% and 46.57%,and It was composed of several monosaccharides including fucose,galactose,galactose,glucose,xylose,mannose and glucuronic acid.By partial acid hydrolysis,periodate oxidation,Smith degradation and nuclear magnetic resonance(NMR),it was found that the main chain of FSF-2 was composed of glucose and xylose,monosaccharide connection way is 1?2,1?or 1?6.(3)Study on the moisturizing activity in vitro found that the hygroscopicity(8h)of CEFSF was better than that of glycerin,butanediol,and sodium alginate,and the moisture retention capacity of CEFSF aqueous solution was better than that of butanediol and sodium alginate,which was equal to 5% glycerin,under relative humidity of 43% and 81%;After purification,the hygroscopicity of FSF-1 was decreased,and its hygroscopicity(8 h)was just better than that of sodium alginate,which was lower than that of CEFSF and butanediol.But the moisture retention activity of FSF-1 enhanced,which was better than that of CEFSF,and less than that of FSF-2;the hygroscopicity(either 8h or 60h)and the moisture retention activity of FSF were further enhanced,which were better than CEFSF and FSF-1.Study on the moisturizing activity in vivo found that 5%CEFSF and 1% FSF-2 can significantly increase the skin moisture content,which were equal to the effect of glycerol,and the ability that reduce the percutaneous water loss amount of them were significantly better than glycerol,can significantly reduce the percutaneous water loss amount.(4)With moisturizing activity as the index,the optimal ratio of glycerol and CEFSFwas selected as follows: the concentration of glycerol was 3% and the concentration of CEFSF was 2%.Design a skin cream in which glycerol and CEFSF were added according to the optimal mixture ratio,and prepare three control skin cream.The physical and chemical properties of the four cream and the heat resistance and cold resistance are in accordance with industry standards;The results of skin test showed that: compared with the control cream,the cream added with CEFSF-glycerol misture could significantly increase the skin moisture content,and can effectively inhibit the skin water loss.
