Ginseng, with high pharmaceutical value in traditional herb materials in China, is attracting more and more scientist’s attention. One of the study branch is to detect the active ingredient: ginsenosides in ginseng and its compound preparation, the main detection methods including high performance liquid chromatography(HPLC), high performance liquid chromatography mass spectrometry(HPLC-MS), nuclear magnetic resonance spectroscopy(NMP) and so on. However, such methods has a one common shortcoming: it needs extracting, separating,enrichment section or some complex sample pretreatment, the whole detecting process is time-consuming and poor efficiency.Ambient ionization mass spectrometry is a new technique that can detect complex constituents without any sample pretreatments or some simple preteatments, largely improved the efficient of sample analysis. In this study we used ginseng and its Chinese patent drugs as studying objects, using ambient ionization mass spectrometry to rapid detect its ginsenosides and using this way to detect active ingredients in different forms of the drug. The main contents are as followed:1. Illustrate the current situation of Araliaceae and Panax ginseng and emphasize the usage of Chinese patent drugs among consumers. Through the comparison of conventional method of drug detection, discuss the advantages of ambient ionization technique in real herb material analysis.2. Under the majorization reaction process of MS detecting ginsenosides, this study used 105 ℃ source temperature, methanol as solvent and 15 μL for each sample injection volume to qualitative and quantitative the ginsenosides in ginseng. Compared the result with that in HPLC-PDA method. It shows that ambient ionization mass spectrometry can semi-quantitative some real herb materials in reasonable concentration and at a certain extent achieved rapid analysis, without secondary pollution and largely improved the usage of samples.3. Compared the spectrum of ginseng sample and its standard compounds, identify the content of ginsenosides in ginseng sample. Select different parts of one ginseng: root,bark,tassel,stem and Chinese patent drugs as studying objects using ambient ionization mass spectrometry to semi-quantitative its ginsenosides.4. Using gravity column chromatography to separate ginsenosides from ginseng extract, using ambient ionization mass spectrometry and thin layer chromatography to detect ginsenosides in different fractions. It shows ambient ionization mass spectrometry can rapid detect each fraction and compared with thin layer chromatography, ambient ionization mass spectrometry with high efficiency, accuracy and can apply in real productive process.
