Ion mobility spectrometry(IMS)is a chemical analysis technique,in which compounds are characterized based on the difference of migration rate of various ions in the gas phase, this technique features advantages of simple operation, rapid respond, sensitive detection, high reliability and low cost,et al.In the early stage, the IMS was mainly used for the rapid detection of explosive, dangerous materials such as drugs and chemical warfare agents. In recent years, with the continuous improvement and development of IMS technology, its application range has rapidly expanded to many areas such as drug analysis, food safety control and environmental pollutant detection. With the appearance of portable IMS instruments,a single analysis can be completed in milliseconds, which makes IMS an irreplaceable means of rapid in-situ screening.Based on the excellent performance of IMS in in-situ rapid screening, this paper studied the application of rapid screening performance of IMS in the detection of illegal ingredients in Chinese traditional medicines or foods(including feed)and in the detection of DUID(Driving Under the Influence of Drug). The main contents are as follows:Part 1: Rapid in-situ screening of five non-steroidal anti-inflammatory drugs illegally added in anti-rheumatic Chinese traditional patent medicines by IMSObjective: To study the IMS migration behavior of five non-steroidal anti-inflammatory drugs, and utilize the IMS method for the rapid in-situ screening of illegal ingredients in anti-rheumatic Chinese patent medicines. Methods: Take five NSAIDs drugs for IMS detection, determine the migration parameters;optimize IMS operating parameters to achieve a optimalresponse sensitivity. The collected anti-rheumatoid Chinese medicine samples were directly dissolved and diluted without any sample pretreatment procedure, then the solution was injected into IMS for analysis. Results: In the positive mode, the five NSAIDs seperated well, good linearity and precision were obtained in the range 0.06-0.33 ?g/mL.After dissolving, the sample solution can be directly injected into the IMS for analysis. The target components are well separated from the matrix with no matrix interference observed. The recovery rate of the method is greater than98.5%.Conclusion: The method is simple and quick in operation.A single analysis can be accomplished in one to two min,as a result, both rapid in-situ screening and rapid qualitative or semi-quantitative analysis of target components can be achieved. Therefore, the ESI-IMS method established in this study is very suitable for rapid screening and judgment of illegal addition of NSAIDs in anti-rheumatic Chinese patent medicines.Part 2: The rapid detection of veterinary drug olaquindox illegally added in pannage by centrifugal ultrafiltration coupled with IMSObjective: To establish a centrifugal ultrafiltration(CF)method for the pretreatment of feed samples, complete the in-situ rapid screening of illegally added olaquindox in swine feed samples by CF coupled with IMS method. Methods: Analyzed the standard solution of OLA by IMS to determine the optimal detection parameters;established a closed centrifugal ultrafiltration device, utilized the CF device for the pretreatment of the collected real feed samples, and then the filtrate was injected into the IMS for the study of illegal addition. Results: Good linearity of OLA was obtained in the range of 25.0-250?g/mL.The detection limit of the method was up to 0.4 mg/kg.No matrix interference was observed when the real feed sample was analyzed. The method provided a good specificity and the recovery rate was greater than89.6%.The entire measurement process was quick and simple. Conclusion: The method is easy to operate, with a high detectionsensitivity,by utilizing this method, the determination of feed samples can be completed within 11 min without any matrix interference, thus the method can be used as an in-situ rapid screening method for OLA in feed samples.Part 3: Rapid determinetion of chlorpheniramine maleate in saliva samples by IMSObjective: A liquid-liquid extraction-ion mobility spectrometry method was utilized for the rapid detection of chlorphenamine maleate in saliva samples, and intend to provide a rapid screening method for drug driving law enforcement. Methods: In this part,1ml saliva sample was collected, and diluted to 2mL with distilled water, then extracted by ethyl acetate. The extract phase was evaporated to dryness under nitrogen, then reconstituted with 100 ?L of methanol, and inject into IMS for determination. Results: A good response of chlorpheniramine maleate was achieved in IMS,and the detection limit of the method was up to 1.05 ng/mL.No matrix interference was observed in the analysis of QC samples, and the recovery rate of the method was greater than 88.1%.A single analysis can be accomplished in only 6 min. Conclusion: The method established in this study is proved to be simple and rapid.A single analysis can be accomplished in only 6 min, thus the rapid detection of chlorphenamine maleate in saliva samples can be achieved. Therefore, the method established in this study can be used as a method of in-situ screening for drug driving, and provide an immediate in-situ rapid analysis method for drug driving enforcement, thus helping ensure traffic safety.
