Epimedium is one of edible medicinal plants promulgated by the Chinese Ministry of Health (the present National Health and Family Planning Commission of China). Epimedium plays important roles in food industry. It is useful to discuss the quality control of Epimedium and to detect nutrients and active substances in Epimedium. The quantitatively analytical method based upon near-infrared spectroscopy is proved to be a rapid, precise, accurate and non-destructive alternative to the conventional technique. In the present dissertation, near-infrared diffuse reflectance spectroscopy technique was applied to the determinationof moisture, ash, flavonoids, polysaccharides and alkaloids, which may provides a basis for quality control of Epimedium and development of healthy food containing Epimedium.A total of 130 batches Epimedium samples were collected from different areas of China and 8 important quality indicators (i.e. moisture, ash, polysaccharides, epimedin A, epimedin B, epimedin C, icariin and alkaloids) were analyzed. This is the first study to establish the model of the water ash, polysaccharides, epimedin A, B and alkaloids by based near infrared diffuse reflectance spectroscopy, and used the new method of quantitative analysis to dectect the Epimedium sample content related nutrients and active substances. The steps of near infrared diffuse reflectance spectroscopy method as follows:The first step is the use of conventional chemical test method for the determination of all samples 8 quality index value of laboratory analysis; the second step is to use the near-infrared spectroscopy instrument for all sample spectra collection; The third step is combine the laboratory analysis value with near-infrared spectra by the way of mathematical modeling (spectral approach to scattering, mathematical processing parameters and regression analysis)to optimize the impact of the model parameters, select the most optimal parameters of the model results, and establish the near infrared analytical model Epimedium quality control quantitative detection; The fourth step is to validate the model and the actual sample test to examine the merits and feasibility of the model. The near infrared model of moisture, ash, epimedin A polysaccharides and alkaloids are using "SNV+D" scattering approach, 2,4,4,1 "mathematical approach, and "MPLS" regression analysis, these types of treatments to get the model have the good effect, the scaling coefficient of determination (RSQ) were as follows:0.9387,0.8843,0.8962,0.7782,0.8997; the cross-validation correlation coefficient (1-VR) were:0.8519,0.8009, 0.8754,0.7829,0.8457; The near infrared model of epimedin B which uses "IMSC" scattering approach, using "1,8,8,1 mathematical approach, and "MPLS" regression analysis,these types of treatments to get the model have the good effect, the scaling coefficient of determination (RSQ) was 0.9357, the cross-validation correlation coefficient (1-VR) was 0.9238; the near infrared model of epimedin C using "Detrend" scattering approach, "2,4,4,1 mathematical approach regression analysis. These types of treatments to get the model have the good effect, the scaling coefficient of determination (RSQ) was 0.9235, the cross-validation correlation coefficient (1-VR) was 0.9173; the model of Epimedium polysaccharide using near infrared model "SNV" scattering approach, "2,4,4,1" mathematical approach, the "MPLS" regression analysis.These types of treatments to get the model to have the good effect, the scaling coefficient of determination (RSQ) was 0.8352, cross-validation correlation coefficient (1-VR) was 0.7941:and external validation is better, the decisions of the eight quality indicators higher coefficient, the error is small, so the scaling effects are good, and reached practical extent, So it can be quickly predicted. On this basis, this study built a new near-infrared diffuse reflectance spectroscopy to detect the moisture, ash, polysaccharides, epimedin A, epimedin B, epimedin C, icariin, and other nutrients and active substances of the Epimedium. The water and ash content of the commercial Epimedium herbs in our country were first measured,the pass rate was 100%,82%, respectively. And the nine different varieties of Epimedium were detect, the results indicate the presence of various indicators content among various species of Epimedium exists big difference, there is a big differences from the the park planting and wild planting Epimedium, but the content of all the indicators are within reasonable limits. A large variation of contents of moisture, ash, polysaccharides, epimedin A, epimedin B, epimedin C, icariin and other active substances was found among these Epimedium samples from different origins. As a result, the future research and development need to focus more on these 8 important quality indicators.Near-infrared diffuse reflectance spectroscopy is a feasible and reliable method for quantitative analysis of moisture, ash, polysaccharides, alkaloids, epimedin A, epimedin B. epimedin C and icariin in Epimedium. The proposed method shows great potential for becoming a new technique for quality control of Epimedium materials and healthy foods containing Epimedium.
