Rhizoma polygonti is mainly distributed in Fujian, Zhejiang and Yunnan. There are three classifications in Fujian:Polygonatum odoratum, Polygonatum filipes and Polygonatum filipes. Meilie area in Sanming city is the main production areas in Fujian. In ordre to develop and use the resource furtherly, extraction technology, Isolation and purified technology and the anti-oxidantive effect of Polygonatum filipes polysaccharide were studied. Therefore, basic data for development of Polygonatum filipes polysaccharide were offered.(1) The effects of various factors on ultrasonic-assisted extraction of Polygonatum filipes polysaccharide were studied. The optimal ultrasonic-assisted extract technology was confirmed. Effects of liquid material ratio, extraction temperature, ultrasonic power and ultrasonic time on extraction rate of polysaccharide from Polygonatum filipes were determined individually. Based on these results, a quadratic regression rotational combinational design was applied to establish a regression model and estimate the optimal technology for maximal extraction yield. The results showed that the optimal ultrasonic-assisted extraction technology were as follows:liquid-solid ratio 17.81:1 (mL:g), ultrasonic time 59.74 min, extraction temperature 72.97℃, ultrasonic power 152.85 W, the yield was 16.6471%. To test the reliability of the method, the yield was 16.59%.(2) Removal of protein from crude polysaccharide of Polygonatum filipes was studied. Hydrochloric acid method, Sevag method and TCA method were used to remove protein from polysaccharides of Polygonatum Filipes. The result showed that hydrochloric acid method deproteinized worst, protein removal rate was only 45.7%, polysaccharide retention rate was 78.6%; though TCA method was good, it had trival operations, and protein removal rate was 75.88%, the retention rate of polysaccharide was 93.55%; Sevag method protein had the best effect with deproteinization rate of 90.94% and polysaccharide retention rate of 81.16%.(3) The process of Polygonatum filipes polysaccharide decolorization was studied in this paper, decolorization treatment on the extraction of Polygonatum filipes polysaccharide by reverse micelle method and macroporous resin adsorption method, the influence of the factors under different process conditions on the decolorization rate and loss rate of polysaccharide. The results showed that the best decolorization effect of Polygonatum filipes polysaccharide is macroporous resin adsorption method. By comparing eight kinds of adsorption resin, NKA-Ⅱ resin is the best, the static adsorption and the optimum conditions were:pH6, bleaching temperature 40℃, bleaching time 4h, resin content is 2g/10mL, the decolorization rate of polysaccharide was 72.35%, the loss rate of polysaccharides was 23.18%; the dynamic adsorption and the optimum conditions were:the flow rate of 2BV/h, the sample volume was 2BV, the concentration of sample is 1mg/mL, the decolorization rate of polysaccharide was 82.47%, the loss rate of polysaccharides was 27.68%.(4)The antioxidation activity of Polygonatum filipes polysaccharide was studied in this paper. Effect of different concentrations of polysaccaride extracted from Polygonatum filipes on fecundity, lipofuscin (LF) and catalase (CAT) of drosophila melanogaster was investigated.The results showed that the treated group all could improve the fecundity of drosophila melaaogaster; the 1.6% group had the best effect to reduce the content of LF in drosophila and increase the CAT activity;They suggested that polysaccharide of Polygonatum filipes could promote antioxidant activity of Drosophila melanogaster.
