Lycopene is a natural pigment contained in plants. Lycopene is a kind of the carotenoids, Lycopene is a kind of safety of natural pigment, without any side effects. Lycopene is one of the strongest antioxidant found in plants. It is helpful of anti-cancer, anti-aging, inhibiting tumor, antioxidation, can enhance the immunity, and have many physiological functions. Because of its unique physiological function, lycopene is known well by people gradually. Compared with chemical synthesis method, the lycopene produced with fungal fermentation is a single structure, the activity is maximized.In addition, the method is not only cost saving, but also reduce the pollution and energy consumption and not restricted by season.The experiment produces lycopene by fermentation of Blakeslea trispora. The optimal fermentation conditions are obtained. Corn flour is selected to be carbon source, bean flour is selected to be nitrogen source, the proportion of positive and negative inoculation with Blakeslea trispora is 1:10, the fermentation temperature is determine to be 26.8℃. The optimal medium pH of substrate is determined to be 7.79, the optimal fermentation time is determined to be 130.03 h. 13-ionone, as fermentation accelerator, is added to zymotic fluid after 24 h, imidazole, as fermentation blocker, is added after 42 h. on this condition, the maximum of lycopene ield is up to 1.297 g/L.Zymotic fluid is pretreated by high-pressure homogenization, the homogeneous power is curtained to be 36-40 MPa, homogeneous cycle is curtained to be 5 times, the material liquid ratio is 1:5 (g/mL). The fully homogeneous process will be repeated three times in order to make the mold of lycopene in wall-breaking. The optimum conditions of extraction are as follows:acetone is selected to be extraction agent, the ratio of materials is 1:200, the leaching temperature is 40℃. The experiments determined the optimal column chromatography conditions,100-200 mesh chromatography on silica gel is selected to be sorbent, the temperature of adsorption and elution is 25 ℃, The sample size is 20 mL, acetone is selected to be eluent, the flow rate of elution is 3.5 mL/min.The experements utilization simulated moving bed as actual operation carrier, The theoretical value is optimized by the experiment. The underbrush parameter of coarse extraction of lycopene are optained. The switch time is 490 s, feeding velocity is 2.51 L/s, flow rate of eluent is 5.27 L/s, extract velocity is 4.26 L/s, extract velocity of liquid core is 3.52 m/s, flow rate of circulating is 7.08 L/s. On this condition, the purity of lycopene is up to 98.41%, the yield of lycopene is up to 80.32%.
