This study screened out the different methods of extracting gardenia components to filter by DPPH radical scavenging capacity, and optimize the subcritical extraction process. The study was also eseparated the different polarity composition of gardenia extract and analyzed it’s constituent analysis and biological activity. The main results were as follows.(1) Study the different methods for screening of active ingredients of the extraction of gardenia ftuit. We compared the different extraction of gardenia from these four methods of reflux, ultrasonic, microwave, and subcritical ethanol by antioxidant activity.The results show that the DPPH radical scavenging capacity of the subcritical ethanol gardenia extracts is the strongest, whose IC50 values of DPPH radical scavenging capacity was 193.90 mg/mL. At the same time, the results of the constituent analysis and the acetyl cholinesterase inhibition activity show that the subcritical ethanol extract is better than other extraction methods.It’s total cycloalkene ether terpenoid substances content is 68.86% and the acetyl cholinesterase inhibition rate was 70.39% at the concentration of 5.0 mg/mL.(2) Study the subcritical extraction process conditions optimization. In terms of single factor experiments, response surface method was designed to study the best condition of extracting by the value of DPPH radical scavenging capacity.Selection of extraction temperature, extraction time, ethanol concentration, three single factor on DPPH radical scavenging ability as the response value, and using the response surface method design three factors three levels, optimized the conditions of subcritical ethanol. The optimum condition was the ethanol concentration of 60%, extraction temperature 140 ℃, extraction time 9 min.(3) Separate the aqueous solution of gardenia subcritical ethanol extract. This study is to separate the subcritical ethanol extract, and obtained four polar fractions, they were petroleum ether, n-butanol, ethyl acetate and water fractions in turn. First of all, we analyzed different component composition. Results showed that the main ingredients of different fraction is cycloalkene ether terpenoids. The results of HPLC show that the most abundant of cycloalkene ether terpenoid is glycosides.The ethylacetate phase has a highest content of cycloalkene ether terpenoid, about 80.95%, and it contents glycosides about 196.09 mg/g.This study determination the activity of each phase. The ethyl acetate phase show the strongest activity of, the value of DPPH radical scavenging ability IC50 is 39.08 mg/mL, and its activity of acetylcholinesterase inhibition rate of 90.64% at the concentration of 1.0 mg/mL.Finally, this study observed the polar phase extract on PC12 cells protection by the model of H2O2 induced injury. The results showed that the polar phase extract has obvious protective effections in the range of concentration of 6.25 ?g/mL to 100 ?g/mL.
