This study was supported by Project of National Key Technology Research and Development Program for The 12 th Five-year Plan(No. 2012BAD33B03). With the increase of population aging and accelerated social pressure, memory loss or degradation has become one of the main problems of troubled people. Therefore, the search has been to improve the memory effect of the substance of the current research focus. At present, people often use animal behavior experiments to assist enhance the research aspects of memory, but there is a large amount of experimental and interference by many factors other defects of this method. Existing research shows that the central nervous system caused by oxidative stress is an important factor in memory decline or degradation, therefore, the present study has assisted in the screening effect of corn peptides enhance memory, a preliminary to the antioxidant capacity index, screened with anti strong oxidation of the peptide to the next step of the experiment.In this study, corn gluten meal as the raw material, the use of biological enzyme, membrane separation and freeze-drying technology to prepare different molecular size of corn peptide; visible light compared different components of corn peptides in vitro free radical scavenging activity of the size of a colorimetric technique; the use of preliminary screening of corn peptide component of PC12 cells to nerve cells were studied; based on this preliminary study corn peptide improving links with learning and memory function between the antioxidant activity; Finally, the development of a maize as auxiliary memory enhancement peptide drinks, corn gluten meal and efficient use of a new way. The main results was as follows:Alcalase alkaline protease after enzymatic hydrolysis of corn protein by ultrafiltration and freeze-dried to obtain molecular weights of <1 k Da, 1 k Da ~3 k Da, 3 k Da ~10 k Da, 10 k Da ~30 k Da four corn peptide component CPS1, CPS2, CPS3, CPS4.In DPPH radical, ABTS radical, hydroxyl radical, superoxide anion radical scavenging and ferrous ion chelating ability as an index, comprehensive evaluation of the CPS 1,CPS 2, CPS3, CPS4 in vitro antioxidant activity. The main results was as follows: Capacity at a concentration of 0.01mg/m L~0.5mg/m L, the components were cleared ABTS radical: CPS1> CPS2> CPS3> CPS4; Capacity at a concentration of 0.1mg/m L~2mg/m L, the components of the DPPH radical were: CPS1> CPS2> CPS3> CPS4; At a concentration of 0.5mg/m L~2mg/m L, the components of superoxide anion, hydroxyl radical capacity in the order of: CPS1> CPS2> CPS3> CPS4, chelated divalent iron ion ability were: CPS2> CPS1> CPS3> CPS4. Whereby the antioxidant activity of selected good components CPS1 and CPS2 for further research.PC12 cells are tumor cells of one kind of the sympathetic nervous system, with its oxidative damage model is established based on an important pharmacological model used to filter the active ingredient in vitro.For initial screening of two components CPS1, CPS2 carried PC12 cell toxicity, the use of MTS cell viability assay, no significant effect(P>0.05) the vitality of PC12 cells showed no cytotoxicity and cell proliferation. Establish injury model in PC12 cells, H2O2-induced PC12 cells were pre-protection, adding 10μg/m L~1000μg/m L concentration of corn peptide CPS1 and CPS2, experimental results show that: the added concentration of 1000μg/m L of CPS1 experimental group increase the cell viability significantly than H2O2 injury group( P<0.01), have the strongest cytoprotective.The high, medium and low concentration of CPS1 experimental sample to male mice as experimental subjects, relying on blind-side water maze test, the shuttle tank experimental and avoid black-box experiment experimental to analysis the learning and memory of mice. The main results was as follows: In water maze test, fed corn peptide CPS1 high-dose group latency of reach the blind end I than the other groups were significant(P <0.05); passive avoidance test, fed corn peptide CPS1 high dose group of mice the number of errors than the other group, the difference was significant(P <0.05); shuttle box test, the average number of errors fed corn peptide CPS1 experimental group of mice the number of errors compared to the control group of mice was not significantly different(P > 0.05), may be electrical stimulation had an impact to mice normal activity. No spontaneous activity frequency with the control group was given high-dose mice compared to corn peptide CPS1 significant difference(P> 0.05), the number of caving and high-dose group was fed corn peptide CPS1 mice had significant difference(P <0.05). It concluded, indicating that high doses of corn peptide CPS1 improve memory in mice of the most prominent.In maize peptide CPS1 meters peptides on the production performance of mice in vivo antioxidant indexes and brain biochemical indicators, the research group compared with normal control group, the body weight of mice, feeding conditions, organ coefficient was no significant difference, growth index changes normal mice. Compared with the control group, the content of SOD in mice fed corn peptide CPS1 experimental group significantly increased than the control group of mice serum SOD content(P <0.01), while the serum level of MDA should significantly lower than the control group of mice serum content of MDA(P <0.01). Compared with the control group, high, medium and low doses of peptide CPS1 maize can be significantly reduced in mice brain Ach E vitality(P<0.01), high dose group of mice brain ACh E content with low-dose group II mouse brain the ACh E content had significant difference(P<0.05), description high doses of corn peptide on mice brain ACh E content will result in a significant impact.Based on the above experimental results on an auxiliary we designed a peptide enhanced memory corn beverage. Sensory evaluation and the use of orthogonal design method developed corn peptide drink recipes: 2% CPS1,70% jujube juice, 0.2% citric acid, 8% xylitol and 0.25% CMC-Na. Detection obtain soluble solids ≥5.0 g / 100 m L, soluble protein ≥3.3%, polysaccharide ≥2.5%, in line with the national standard drinks, it ensure the safety of edible, finally got a functional beverage, its future large-scale production has laid a good foundation.
