Florometric detection has been increasingly used because of its low cost and rapid operation. And reaction-based probes are widely used in food safety, medical diagnosis and environment pollution detection etc, for their advantages of high sensitivity and good selectivity. Taking account into these advantages and the practical significance and urgent requirement of cyanide ion(CN-) and hypochlorous(HClO) detection, we designed and synthesized two novel probes here.A near-infrared probe SY was successfully developed for CN-. SY exhibited an unique colorimetric(A356/A440) in UV-vis, dual-ratiometric(I414/I564 and I803/I564) in fluorescence towards CN- and under various absorption(356 and 440 nm) and emission(414, 564 and 803 nm) channels, offering versatility during measurement. The comparison results show that the ratiometric result is more accurate than the single channel detection. And the minimum detection limit of SY was 5.4×10-8 M, which was far below the maximum level 1.9×10-6 M in drink water by WHO. Furthermore, SY could be used to measure β-glucosidase by detecting CN- traces in commercial amygdalin samples. Overall, the results indicated that SY is a selective sensor for CN- and a potential indicator of β-glucosidase.A novel probe for HClO was developed by oxygenolysis of azo moiety for the first time. Compared with the reported probes for HClO, YDN showed excellent water solubility as well as high selectivity and sensitivity. Its application in cell and vivo imaging would contribute to insight into the HClO formation and inflammation treatment in the future. Convincing evidences had shown that reductive buffer like 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid(HEPES) and tris(hydroxymethyl)aminomethane(Tris) etc. was not suitable for HClO detection, providing valuable reference for the solution selection in other reactive oxygen species(ROS) detection later.
