With the improvement of living standard, people pay more attention to keep in good health, and nutrition health care has become a fashion trend. There are varieties of health-care products in the market, while most of their safety and health care function can not be guaranteed. At present, foods with health care function extracted from the natural ingredients are deeply loved by people. e.g. edible fungi polysaccharides, tea-polysacharides and fortunella margarita polysaccharides, etc. At the same time, Chinese herbal medicine with health care function was also paid more attention by consumers. As a big country in herbal medicine, China not only has a long history, but also has rich species. But the tedious steps in preparing edible Chinese herbal medicine hinder its rapid development. Since joining into WTO, our country has a big increase in exporting Chinese herbal medicine, but all of them are exported in the form of raw and low added value, with lower volume of business than other countries. Therefore, studying on deep processing of Chinese herbal medicine and establishing a safe and reliable Chinese herbal medicine processing system, have the vital significance. Taking Anoectochilus as raw material, using the technologies of heat pump dehydration, ultrahigh pressure extraction and membrane separation, to screen a set of safe and reliable, high efficiency and quality of Anoectochilus polysaccharide extraction process, develop Anoectochilus polysaccharide oral liquid and fill the domestic blank. The experimental results are as follows:(1)With Anoectochilus as raw materials, using natural drying, drying, heat pump dehydration and other drying methods to prepare dried Anoectochilus, and with phenol-sulfuric acid method to measure Anoectochilus polysaccharide content. The results showed:the Anoectochilus polysaccharide content, fresh> heat pump dehydration> drying> natural drying, the yield rate of polysaccharides, are respectively 11.56%,11.01%,9.12% and 8.34%. After membrane treatment, the loss rates of Anoectochilus Polysacchride are different, fresh> drying> heat pump dehydration> natural drying. Relative to other drying methods, the yield rate of Anoectochilus polysaccharide by the heat pump dehydration is the highest, and drying time is the shortest, after membrane ultrafiltration, the loss rate is the lowest, and the Anoectochilus Polysacchride product has good color and high quality.(2) Using Anoectochilus dried by heat pump as materials to compare yield rates of Anoectochilus polysaccharides by hot water extraction method and ultra-high pressure assisted extraction method, and optimize its extraction parameters.. Selecting the temperature, time, ratio of liquid to solid as single factor of the hot water extraction method, and optimize the data with the response surface experiment. Choosing the operating pressure, hold-up pressure time, temperature, ratio of liquid to solid as single factor of Ultra-high pressure assisted extraction method, Using the orthogonal test method to design experiment and range and variance analysis to handle the experimental data.The results showed that the optimum extracting process parameters of Anoectochilus by hot water extraction:extracting temperature 82 ℃, extracting time 1h, ratio of liquid to solid 30:1, and the yield rate of polysaccharide was 7.33%.The optimum extracting process parameters of Anoectochilus by ultra-high pressure assisted extraction:operating pressure 300Mpa, the ratio of liquid to solid 1:30, hold-up pressure time 20min, extraction temperature 50 ℃. and,the yield rate of polysaccharide was 12.19%. The analysis results showed that the ultra-high pressure assisted extraction method can effectively improve the yield rate of polysaccharides.(3) Using secondary ultrafiltration to purify Anoectochilus polysaccharide. Anoectochilus polysaccharide extracting solution is classified by 100kDa ultrafiltration, operating pressure is 0.4Mpa; then using lOkDa ultrafiltration to grade the permeated liquid, operating pressure is 0.3Mpa, after the purification, the collected solution is polysaccharide Anoectochilus solution, relative molecular weight is between 104 to 105. Compared with other polysaccharide purification methods, ultrafiltration, which can treat with multitudinous sample liquid with low cost, is suitable for industrial production.(4) Development of Anoectochilus polysaccharide oral liquid. The polysaccharides oral liquid mainly includes the purified Anoectochilus polysaccharide solution, and excipients are honey, white sugar and citric acid. We have developed a nutritious, safe and delicious oral liquid. The optimal formula is as follows:the contents of Anoectochilus Polysaccharide, white sugar, honey and citric acid are 1.6%,4%,1% and 0.07%, respectively.
