Rosa davnrica Pall. is a kind of promising wild economic plants, Rosa fruitage has a high medicinal value, Rosa seeds also contain rich nutrients, such as Rosa oil (8 wt/wt%).However, as a manufacturer of waste, Rosa seeds are generally not used in the country, leading to irrational resource consumption.Sometimes Rosa seeds are resulting in a waste of resources.Therefore, if we can make full use of the Rosa seed oil and procyanidins, changing the Rosa seed from waste into treasure for Rosa development industry has brought enormous economic benefits.Rosa davnrica pall seeds from the Changbai Mountain were used as raw material to extract Rosa seed oil by ultrasonic assisted method in this study.Using defatted Rosa seed extract proanthocyanidins by microwave assisted method.And Rosa seed proanthocyanidins crude extract was purified by the best models of macroporous res in.Finally, research the lipid-lowering effect of Rosa oil and procyanidins through animal experiments.The main contents and conclusions are as follows:(1) Referring to the national standard, measured Rosa seed crude fat content was 12.40% by Soxhlet extraction method;Measured Rosa seed moisture content was 8.49% by direct drying method;Measured Rosa seed content of procyanidins was 1.55% by iron catalyzed method.(2) Based on single factor experiments, optimized Rosa oil and procyanidins extraction process by response surface analysis experiments.The mathematical model fit was good.Select hexane as the extraction solvent, determined the optimal Rosa oil extraction conditions were:grinding particle size was 60 mesh, solid-liquid ratio was 1:4, ultrasonic temperature was 50℃, ultrasonic power was 200 W, ultrasonic time was 30min, extraction times was 2 times, Rosa oil yield was 11.92%.Rosa oil quality was better by this extraction method.Determined the optimum extraction of the rosa procyanidins as follows:extracting time 70 s, the ratio of material to liquid ratio 1:20, microwave power 360 W, ethanol concentration 60%. The proof test gained the PC yield of the single extract was 72.58%.When the extraction times were 3, the Rosa seed PC yield could reach 93.19% .Purified the crude proanthocyanidin extract with the DM-21 macroporous resin.Eluent was 40% ethanol.After purification, the purity of Rosa seed proanthocyanidins was 63.28%, increased by 6 times compared with unpurified.(3) Kunming male mice were chosen to establish mouse model of hyperlipidemia by high fat diet (cholesterol, egg yolk powder, lard) feeding.We studied the influence of Rosa oil and proanthocyanidins on hyperlipidemia mice.Gavaged five weeks and measured indicators in mice serum and liver tissue.It was found that, compared with the model group, Rosa seed oil and proanthocyanidins no significant effect on body weight in mice and major organs index. It showed that both cound not hurt the animal’s body (liver, kidney, heart and other major organs).Moderate doses (100 mg/kg), high dose (200 mg/kg) of Rosa seed proanthocyanidins could significantly reduce fat mouse serum LDL-C, TC and TG levels (p<0.05), increased significantly mouse in liver tissue SOD, CAT, GSH-PX content (p<0.05), reduced significantly MDAcontent in liver tissue.For effecttion of HDL-C, TNF-a and IL-6 levels, the difference was not significant.Low dose (80 mg/kg) Rosa oil could significantly reduce TG levels in hyper lip id em ia mouse serum (p<0.05) and significantly increased CAT content in liver tissue (p<0.05).The influence of other indicators was not significant.lt proved that low doses of Rosa seed oil and high doses of Rosa seed proanthocyanidins had a lipid-lowering effect on hyperlipidemic mice.
