| Cantharelles cibarius is a kind of outer root fungus, which is rich in polysaccharides, amino acids, trace elements and other nutrients. The modern pharmacological research indicated that Cantharelles polysaccharides obviously reduce blood pressure, have anti-tumor activity. In addition, Cantharelles polysaccharides also has oxidation resistance, and can remove excess free radicals in the body, which is a natural nontoxic antioxidant. At present, there were few reports on Cantharelles polysaccharides preparation. The present paper was based on the modern pharmacology findings and the modern technology, Cantharelles polysaccharide oral liquid was prepared by extracting the effective component of Cantharelles.This topic has completed the following work:1. In the extraction process research, taking the polysaccharide content as the target,the extraction process was investigated by the single factor experiment and the orthogonal experiment. The experimental results showed that the optimal experimental parameters were 30 minutes ultrasonic, 50℃extraction temperature, 1:30 ratio of material to water, 2 hour extraction time, polysaccharide yield was 10.12%.2. In the Cantharelles polysaccharide depuration process, the topic adopted calcium chloride and activated carbon to purify the Cantharellus polysaccharide. The best craft of calcium chloride method was: extract of Chanterelles polysaccharides were added 5% calcium chloride, then heated to 85℃, with the mixing speed being 100 r/min.The best decolorizing-process of the Cantharellus extraction was that stirring at 30℃for 50 minutes with 1% of the active carbon.3. In the Cantharellus polysaccharide oral liquid preparation research, pH,preservative types, corrigent concentration was investigated. By comparing the senses and the clarity, the formulation of Chanterelles polysaccharides oral liquid was: extract of Chanterelles polysaccharides were adde 0.4% citric acid and 0.2% benzoic acid, pH value was styed 4.0-5.0 with dilute hydrochloric acid.4. In the Cantharellus polysaccharide oral liquid oxidation resistance research,through restore research, use of(·OH) generated by Fenton system, antioxidant activity of Cantharellus polysaccharides was studied. The results showed that Chanterellespolysaccharides oral liquid compared with Vitamine C, the reducing power and the hydroxyl radical scavenging ability was relatively weak.5. In the quality specification research, the topic has established the Cantharellus polysaccharide content determination method. Cantharelles polysaccharides content was determined by phenol-sulphuric acid method. The results showed that The regression equation of the glucose standard curve was obtained as A=9.6075C+0.0669(R2=0.999), exhibiting a good linear relationship within the rang of0.009~0.074 mg.mL-1. The precision, stability and repeatability were good(RSD=0.72%,0.56%, 0.91%, respectively), the recovery was 98.0%, RSD=1.42%. Chanterelles polysaccharides oral liquid has carried on the preliminary stable inspection to the sample stability. The results showed that the stability of the product was good within half a year.The findings indicated that the oral preparation craft of Cantharellus polysaccharides was science and feasible; the quality was stable, was controllable. |
