So far, more than 20 years of market-oriented reforms, so that melamine as a very wide range of industrial applications of chemical raw materials,Its molecule contains 66.6% of the amount of nitrogen, many unscrupulous companies in order to achieve a lower cost of dairy products, milk,feed protein content of the upgrade,Melamine as an additive to the dairy products, feed, etc., to produce a higher protein products.But as an industrial raw material, the human body can not be effectively decomposed melamine, when excessive melamine into the human body, it will inevitably lead to a very large impact on people’s health.At present, for dairy products of melamine determination method is very much, including the detection method, spectral method, nano gold probe method, colorimetric method, but the many method is restricted to conditions, cost, time and other factors, unable to effectively meet the live fast, simple operation, accurate measurement requirements.In view of this, this study based on the relevant research theory, proposed two kinds of melamine detection methods, and through the experiment, the following conclusions are drawn.1.Method for rapid determination of melamine in dairy products by colorimetric method:Gold nanoparticles were fabricated by three sodium citrate reduction method, and the particle size of20 nm was obtained,It can be observed that the color of gold nanoparticles gradually changed from red to blue, then it can be judged by the naked eye to determine whether the dairy products contain melamine.The method has the lowest detection limit of 0.5ppm, and the quantitative analysis of melamine by using spectrophotometric method, found that when the concentration of melamine in the range of 0.5-10ppm;Gold nanoparticles with the best detection time of 2 minutes. The method is simple and accurate.2.Melamine colloidal gold immune chromatography quantitative, qualitative detection test paper:(1) the detection range and sensitivity, the sensitivity of melamine colloidal gold strip was0.5ppm, when the concentrations of melamine than 0.5ppm, test paper detection line t does not appear red, but when the concentration is lower than 0.5ppm determination of melamine found its concentration is low, t shows the color of the deep. Melamine colloidal gold detection test paper for the detection range of 0.25-6ppm.(2) stability: in the 37 C environment, the colloidal gold immune chromatography test strip can be stored for 3 weeks, the shelf life of 2 years.(3) repeatability: 10 parallel tests showed good reproducibility.3.Taking milk powder as an example, the results of gold nanoparticle colorimetry and liquid chromatography were compared and analyzed:The concentration range of gold nanoparticles is higher than that of HPLC, and will not be affected by other substances in milk powder;Averagerecovery rate and RSD were higher than HPLC;HPLC only found that 1 kinds of milk powder containing melamine, for low levels of milk powder is difficult to measure, and the results of the low.
