This paper is based on the pilot demonstration production line of procyanidins from larch barks which was collaborate built by my research group and Yakeshi forest-chemical products co., LTD. And it combined with the actual production situation of procyanidins product process to optimize the purification process using single factor research method. The target is that factory can produce procyanidins, purity> 90%(UV test result), using the optimized purification process. Besides, this paper use "Infrared spectrum three-level identification method" (a combined detection analysis method with one-dimensional infrared spectra, the second derivative spectra and two-dimensional correlation spectroscopy) which was developed by professor Suqin Sun tsinghua university to analyze the procyanidins production process.First, the research which is based on the original purification data and conclusions of larch bark procyanidins gets single factor purification process optimization results are as follows:the best purification process conditions for the sample liquid concentration is 10 mg/mL, the injection fluid velocity is 5 BV/h, eluent ethanol volumefraction is 70%, elution velocity is 5 BV/h. And the factory can steady product procyanidins which purity is higher than 90%, according to this set of optimized purification process.In addition, using the method of "Infrared spectrum three-level identification method", this paper analyze the intermediate procyanidins products during the process of crushing, degreasing, extraction, concentration, purification and drying, with conjoint analysis method by infrared one-dimensional spectrum, second derivative spectrum and two-dimensional correlation spectrum. The analysis results are as follows:in grinding, a large number of long-chain molecules like cellulose and hemicellulose in larch barks is damaged, cause the active substances in larch barks are are more easily to be extracted; in degreasing, the composition types and proportions of barks have little change, the degreasing process has little affect on the larch bark; using ethanol to extract barks which are crushed and degreased, the large proportion of procyanidins are extracted, and the products’ infrared spectra peaks in three IR analysis types are similar to the standarding-procyanidins; in purification, using macroporous adsorption resin to improve the purity of procyanidins products with its purification ability of procyanidins; comparing with oven drying and spray drying process found that the products are produced by two methods has little different in its infrared spectra, and both methods have no effect on the components of products, so both methods can be used in the production process.In conclusion, this paper achieve the purpose of understand the components content change in each production process and discuss the meaning of every production process.
