Traditional Chinese Medicine is a complex system. The composition will be more complex after subjected with a series of metabolic processes or complicated chemical reactions in vivo. UHPLC-MS/MS possesses significant advantages including fast analysis speed, strong selectivity. The research for pharmacokinetic study of multiple effective constituents of traditional Chinese medicines in rat plasma by UHPLC-MS/MS was established. It might provide valuable reference for the efficacy of Traditional Chinese Medicine, prescription principle, mechanism of action, the new preparation of Traditional Chinese Medicine research and to promote the reasonable use of clinical drugs.1. A rapid and sensitive assay based on UHPLC-MS/MS was established for the simultaneous determination of eight components in rat plasma using catechin and daidzein as IS. The separation of eight constituents was achieved with gradient elution using methanol and 0.2% acetic acid aqueous solution as the mobile phase and detected by MRM using ESI source in the positive-negative ionization mode. The concentration range for eight components were 10-6000,1.5-3000,10-15000,2-3600,1.5-3600,4-6000, 2-9000, and 20-18000 ng·mL-1 respectively. Intra-run and inter-run precision were in the range of 1.0%-14.2% and 2.8%-12.9%, respectively. This analytical method was successfully applied to investigate the pharmacokinetics of eight ingredients in rats after oral administration of Hypericum japonicum Thunb extract.2. Qian hu, the dried roots of Peucedanum praeruptorum Dunn. In this study, a selective and sensitive method was developed and validated for the simultaneous determination of ten coumarins in rat plasma. Coumarin and cryptotanshinone were used as the internal standards. Ten analytes were separated by a gradient elution within a runtime of 20 min. The intra-and inter-day precision were less than 13.4%. The sensitive method was fully validated and successfully applied to the pharmacokinetic study of ten coumarins in rats following oral administration of Peucedanum praeruptorum Dunn extract.3. This paper was designed to establish a simultaneous quantitative method of eight constituents in rat plasma. Furthermore, the pharmacokinetics of those components after administration of Plantago asiatica L extract was executed. Chromatographic separation was achieved using gradient elution with the mobile phase consisting of methanol and water (containing 0.15% formic acid) at a flow rate of 0.3 mL/min. The validated method showed good linearity over a wide concentration range, and the LLOQ of eight components were 0.1,35,10, 8,10,15,5 and 5 ng·mL-1 respectively. The extraction recovery of those analytes ranged from 77.8% to 114.6%. The method has been successfully applied to pharmacokinetic study after administration of Plantago asiatica L extract. These results might be helpful for Plantago asiatica L clinical reasonable application and further studies. |
