In order to investigate the feasibility of tissue engineering anterior cruciate ligament constructed by silk fibroin powder blending with polycaprolactone and polyving akohol, fibrous membranes of silk fibroin powder blending with polycaprolactone was made by electrostatic spinning. The properties of the blended fiber membranes were characterized by scanning electron microscope, hydrophilic angle tester, tensile tester, infrared spectrometer and so on. Morever, we studied the in vitro release phenomenon of 5-coated in the blend fiber. Then fibrous membranes of silk fibroin powder blending with polyving akohol wasmade by electrostatic spinning. Esterification crosslinking and crosslinking of glutaraldehyde steam was carried out to improve the properties of the fiber membranes. The specific research contents are as follows:(1) Different concentrations of polycaprolactone nano fiber membranewas made by electrostatic spinning. Morphology and properties was characterized by scanning electron microscopy, hydrophilic angle test and tensile test. The surface contact angle is 96.3 degrees and fiber diameter reaches 380 nm when the concentration of polycaprolactone was 12%. And at this point the breaking tensile strength reaches 8.21 MPa, and the elongation at break of the fiber film reaches the maximum of 70%. So the optimum spinning concentration of a single component is 12 wt%.(2) Silk fibroin powder blending polycaprolactone nanofiber membrane was made by electrostatic spinning. We prepared a set of different silk fibroin powder ratio of nano fiber membrane by adjusting the relative content of silk fibroin powder.The properties of the fiber membranes were analyzed. The adding of silk fibroin powder reduces the thermal stability of membrane. Adding a small amount of silk powder can improve the hydrophilic performance of the membrane, and it will increase the fracture tensile strength of fiber. When the content of silk fibroin is too high, the thermal stability and the tensile strength of the fiber are decreased. Silk fibroin powder does not dissolve in the process of blending and retains its own natural characteristics. The blending fiber membrane has great research value.(3) Silk fibroin powder blending polyvinyl alcohol nanofiber membrane was made by electrostatic spinning, before electrostatic spinning, the performance of polyvinyl alcohol was improved by freezing and thawing cycles of the spinning solution. Esterification and crosslinking modification of the fiber membrane and glutaraldehyde vapor crosslinking modification were used to improve the water resistance of the fiber membrane and retaining sufficient mechanical strength of fiber membrane in wet state. However, due to the time of esterification crosslinking, the water resistance is improved, thermal stability and the mechanical properties of the fiber membrane are decreased. Morever the mechanical strength of fiber membrane in wet state is very low. In the experiment of steam crosslinking of glutaraldehyde, the hydrolysis resistance of the fiber membrane was increased with the increase of the crosslinking time, but the fiber membranes become very brittle after immersion in deionized water and vacuum drying.We will continue to study to improve the water resistance properties of blend fiber membrane with improving its mechanical properties,and scaffold materials with good mechanical properties and biocompatibility will be prepared in the future time.
