Auricularia auricula(A. auricula) is a non-toxic edible mushroom widely used in traditional Chinese cuisine and is known for their pharmaceutical effects in Chinese herbal medicine. A. auricula-judae belongs to the heterobasidiae of the basidiomycete family and is known as a highly nutritious and officinal edible fungus with a high content of carbohydrates and protein. In this study, the extraction of crude polysaccharide(AARP) from A. auricula root, intended to provide the basis for the further development and utilization of AARP. A. auricula polysaccharide(AAP) has a variety of biological activity, using the biological activities of AAP, add it to the malt to product beer with better feeling and has health care effect, so as to realize the deep processing and utilization of A. auricula.A water-soluble AARP was isolated by hot-water extraction and 80%(v/v) ethanol precipitation. Based on the calibration with Dextran, AARP had a molecular weight of approximately 8.6×105 Da. The quantification of monosaccharides of AARP was analyzed using high-performance liquid chromatography. According to the corresponding relationship of peak and migration time, AARP mainly consisted of six kinds of monosaccharides apparently. The mole ratio of mannose, rhamnose, glucose, galactose, xylose and fucose of AARP were calculated as 1:1.64:23.22:1.28:7.35:0.45. Fourier-transform infrared spectroscopy and 13 C nuclear magnetic resonance spectroscopy were used to analyze the structure of the AARP. Based on the data, AARP has a β- glycosidic bond type configuration.AARP is separated using hydrophobic chromatography on an ME-1 resin column. Three major polysaccharide fractions, AARP1, AARP2 and AARP3, were analyzed by Fourier-transform infrared and 13 C nuclear magnetic resonance spectroscopy.The results of antioxidation test in vitro showed that AARP and AARP1 have certain ABTS+ radical scavenging activity, DPPH radical scavenging activity and reducing power. And removal of the ability of DPPH free radicals is best. Because of a relatively small amount of fractions AARP2 and AARP3,therefore no antioxidant activity were studied on these two fractions. The protective effect of AARP on acute carbon tetrachloride-induced liver injury in mice was investigated. Compared with modle control group, treatment with AARP decreased serum aspartate aminotransferase(AST), serum alanine aminotransferase(ALT), and malondialdehyde aldehyde(MDA) levels; increased superoxide dismutase(SOD) activity; and improved hepatic injury in the acute model of liver injury in mice. These results suggest that AARP possesses potent hepatoprotective activity.On the basis of the traditional beer fermentation, AARP which has a variety of biological activity was added to the beer fermentation process as accessories.The results show that the fermentation beer of tank 2 containing 0.05% AAP have the best fermentation effect. Add different amounts of AAP in the fermentation process have little effect on the content of alcohol and diacetyl content of the finished beer. Brewed beer containing A. auricula polysaccharide keeps the traditional beer flavor and the health care function of human body are obtained.
